HPLC法同时测定绿衣枳壳中水合橘皮内酯、橘皮内酯、马尔敏和葡萄内酯的含量 被引量:8

Simultaneous determination of meranzin hydrate,meranzin,marmin and aurapten in Fructus ponciri trifoliatae by HPLC

摘要 目的:建立绿衣枳壳中水合橘皮内酯、橘皮内酯、马尔敏和葡萄内酯同时测定的HPLC方法。方法:采用Diamonsil C_(18)柱(150 mm×4.6 mm,5μm),流动相:甲醇-水(38∶62),流速1.0 m L·min^(-1),检测波长:324 nm,柱温:25℃。结果:水合橘皮内、橘皮内酯、马尔敏和葡萄内酯分别在0.003 8~0.022 9μg(r=0.999 7)、0.005 8~0.034 7μg(r=0.999 5),0.006 5~0.038 9μg(r=0.999 2)和0.822 6~4.935 7μg(r=0.999 8)的进样量范围内呈良好的线性关系;其平均回收率(n=6)分别为103.50%,97.80%,99.51%和105.00%,其RSD值分别为2.1%,1.7%,2.9%和2.5%。结论:本法简便、准确,可用于绿衣枳壳中水合橘皮内酯、橘皮内酯、马尔敏和葡萄内酯的含量测定。 Objective: To develop an HPLC method for simultaneous determination of meranzin hydrate, meranzin, marmin and aurapten in Fructus Ponciri trifoliatae. Methods: The HPLC system consisted of a Diamonsil C18 column( 150 mm× 4.6 mm,5 μm)and the mobile phase consisted of methanol-water eluted in gradient mode (38: 62) at the flow rate of 1.0 mL min-1. The detection wavelength was 324 nm and the column temperature was 25 ℃. Results: The calibration curves of meranzin hydrate, meranzin, marmin, and aurapte had good linear relationship in the ranges of 0. 003 8 - 0. 022 9 μg ( r = 0. 999 7 ), 0. 005 8 - 0. 034 7 μg( r = 0. 999 5 ), 0. 006 5 0. 038 9μg( r = 0. 999 2) and 0. 822 6 - 4. 935 7 μg( r = 0. 999 8), respectively, and the average recoveries ( n = 6) of the four components were 103.5% with RSD of 2.1% , 97.80% with RSD of 1.7% , 99.51% with RSD of 2.9% , and 105.00% with RSD of 2.5% , respectively. Conclusion: According to the methodology validation, this method can be applied to the determination of meranzin hydrate, meranzin, marmin and aurapten in Fructus ponciri trifoliatae.
