miR-133b与miR-155在非小细胞肺癌患者肿瘤及周围组织中的表达及意义 被引量:10

Expression and Significance of miR-133b and miR-155 in Tumor and Its Surrounding Tissues of Patients with Non Small Cell Lung Cancer

摘要 探讨miR-133b与miR-155在非小细胞肺癌患者肿瘤及周围组织中的表达及意义。选择在湖北科技学院附属第一医院接受治疗的47例非小细胞肺癌患者作为研究对象,收集其肺癌组织和周围正常组织标本,采用Real-time PCR检测miR-133b和miR-155在肿瘤及周围组织中的表达情况,分析其与患者临床和病理特征之间的相关性。结果显示,肿瘤组织中miR-133b相对表达量为221.4±1.013,周围正常组织为605.8±1.001,两者比较差异显著(p<0.05);肿瘤组织中miR-155相对表达量为643.2±1.118,周围组织为386.9±1.097,两者比较差异显著(p<0.05)。即Real-time PCR检测患者肿瘤组织中的miR-133表达明显低于周围正常组织,miR-155的表达明显高于周围正常组织;miR-133b和miR-155的表达与患者年龄、性别、病理类型无明显相关性;而与临床分期和淋巴结转移情况明显相关(p<0.05),其中miR-133b表达水平与淋巴结转移呈明显负相关,miR-155表达水平与淋巴结转移呈明显正相关;miR-133b的表达还与肿瘤分化程度明显相关(p<0.05)。综上所述,miR-133b和miR-155的异常表达与非小细胞肺癌的发生和发展密切相关,对二者表达水平进行检测有助于及早发现、筛选和诊断非小细胞癌患者,对患者预后评估具有一定临床价值。 To investigate the expression and significance ofmiR-133b and miR-155 in the tumor and surrounding tissues of patients with non-small cell lung cancer, 47 cases of non-small cell lung cancer patients treated in the First Affiliated Hospital of Hubei University of Science and Technology were selected as the research object. The lung tissue and surrounding normal tissue samples were collected, the expressions of miR-133b and miR-155 in tumor and surrounding tissues were detected by Real-time PCR, and the correlation between the clinical and pathological features of the patients were analyzed. The results showed that the relative expression of miR-133b in tumor tissues and surrounding normal tissues was 221.4~1.013 and 605.8±1.001, respectively, and the difference was significant (p 〈0.05); in addition, the relative expression of miR-155 in tumor tissue and surrounding tissues was 643.2± 1.118 and 386.9± 1.097, and the difference was significant (p〈0.05). After the detection of Real-time PCR, the expression of miR-133 in tumor tissue was significantly lower than that in the surrounding normal tis sues, while the expression of miR-155 in tumor tissue was significantly higher than that in the surrounding normal tissues. The expressions of miR-133b and miR-155 were not obviously related with age, gender, and pathological type of patients, but they were related with the clinical stage and lymph node metastasis (p 〈0.05). The expression level of miR-133b was negatively correlated with lymph node metastasis while the expression level of miR-155 was positively correlated with lymph node metastasis. The expression of miR-133b was significantly correlated with tumor differentiation (p 〈0.05). Besides, the expression of miR- 133b and miR- 155 was closely related with the occurrence and development of non-small cell lung cancer. The expressing detection of them was helpful to early discovery, screening and diagnosis of patients with non-small cell carcinoma, and it had a certain clinical value for prognosis.
