G蛋白调节因子2(G protein regulating factors2,RGS2)表达水平的变化与多种肿瘤的发生、发展有密切关系。为了解RGS2蛋白表达水平与乳腺导管内增生性病变(intraductal proliferative lesions,IDPLs)的关系。应用免疫组化对RGS2,CK5/6进行染色,观察其在98例IDPLs(15例普通型导管增生,11例非典型导管增生,72例导管原位癌)和正常乳腺组织的表达情况。结果显示,在普通型导管增生中RGS2的表达水平分布不一致,而在其余各型的IDPLs中RGS2蛋白的阳性表达呈均匀性分布;CK5/6蛋白的阳性表达只存在于普通型导管增生中,而其余各型IDPLs均不表达;在IDPLs中RGS2的表达水平与CK5/6的表达水平呈负相关性(r=-0.995,p=0.000);随着IDPLs的级别增高,RGS2的表达逐渐丢失。结果表明,RGS2与CK5/6的联合应用可用于对IDPLs的鉴别诊断;RGS2可能作为检测IDPLs生物学行为和判断IDPLs患者预后的一项重要指标。
The expression level change of G protein regulating factors 2 (RGS2) is closely related to the occurrence and development of a variety of malignant tumors. To investigate the relationship between RGS2 protein expression level and intraductal proliferative lesions (IDPLs) of the breast, the immunohistochemical staining of RGS2 and CK5/6 was performed on 98 cases of IDPLs (including 15 usual ductal hyperplasia, 11 atypical ductal hyperplasia, 72 Ductal carcinoma in situ) and normal breast tissues to observe the expression of RGS2 in all IDPLs. Results showed that RGS2 expression was inconsistent in UDH, but it was consistent in other subtypes of IDPLs. CK5/6 positive expression was only detected in UDH but not in other subtypes of IDPLs. The expressive levels of RGS2 protein and CK5/6 protein were negatively correlated in IDPLs (r=-0.995, p=0.000). The expression of RGS2 lost as the grade of IDPLs increased. In a word, the combination application of CK5/6 and RGS2 immunostaining were useful for differential diagnosis of IDPLs of the breast; RGS2 protein might be an important biomarker to evaluate the biological behavior of IDPLs, and to estimate the prognosis of patients with IDPLs.
