摘要
目的:探讨云南汉族人群活化性受体NKG2D基因单核苷酸多态性(SNP)与非小细胞肺癌的相关性。方法:选择云南汉族非小细胞肺癌患者467例(肺癌Ⅰ+Ⅱ期组151例,肺癌Ⅲ+Ⅳ期组316例)和健康对照384例(对照组),采用Taq Man探针基因分型方法对NKG2D基因的2个SNPs(rs2255336和rs2617160)位点进行基因分型,研究其等位基因、基因型及所构建的单倍型在肺癌Ⅰ+Ⅱ期组、Ⅲ+Ⅳ期及对照组人群中的分布差异。结果:rs2255336和rs2617160位点的等位基因及基因型在肺癌Ⅰ+Ⅱ期组、肺癌Ⅲ+Ⅳ期组和对照组中的分布频率的差异无统计学意义(P>0.05);遗传模式分析显示,rs2255336和rs2617160在Ⅰ+Ⅱ期肺癌组和Ⅲ+Ⅳ期肺癌组最优的遗传模式均为Recessive模式,其中CC-CT基因型相对于TT基因型可能是肺癌发展的保护性因素(P=0.040,OR=0.26,95%CI:0.06-1.17);rs2617160位点TT-AT基因型相对于AA基因型可能是肺癌发展的保护性因素(P=0.045,OR=0.60,95%CI:0.36-1.00)。结论:NKG2D基因rs2255336和rs2617160 SNPs位点可能与云南汉族人群非小细胞肺癌的发展有关。
Objective: To investigate the correlation of the singe nucleotide polymorphisms (SNP) of natural-killer group 2, member D (NKG2D) gene with non-small cell lung cancer (NSCLC) in a Yunnan Han population. Method: TaqMan asssy was used to study genotype 2 SNPs (rs2255336 rs2617160) in NKG2D gene in 467 NSCLC patients (including patients in I + Ⅱ and Ⅲ+Ⅳ stages, namely group I + Ⅱ and group m + IV ) and 490 healthy people (control group). The distribution of alleles, genotypes and the constructed haplotypes were analyzed. Results: The allelic and genotypicfrequencies of the 2 SNPs were not significantly different between I + Ⅱ, and control group, and be- tween Ⅲ+Ⅳ and control group ( P 〉 0.05 ). While results of inheritance mode analysis showed that the best fit inheritance model for rs2255336 and rs2617160 in 2 lung cancer groups was recessive mod- el. The CC-CT genotype of rs2255336 ( P = 0. 040, OR = 0. 26, 95 % CI: 0. 06 - 1.17 ) and the Ⅲ+Ⅳ genotype of rs2617160 (P =0. 045 ,OR =0.60, 95% CI:O. 36 -1.00) might be the protective factors against NSCLC development. Conclusions: Polymorphisms ( rs2255336 and rs2617160) in NKG2D gene might correlate with NSCLC in this Yunnan Han population.
