矽尘对小鼠肺组织NF-κB p65 mRNA与TGFβ1 mRNA表达的影响 被引量:4

The effects of silica dust on the expression of NF-κB p65 mRNA and TGFβ1 mRNA in mice

摘要 目的探讨矽尘对实验小鼠NF-κB p65mRNA和TGFβ1 mRNA表达的影响。方法取健康昆明种小鼠40只,随机分为4组,每组10只,分别为2 h组(每天吸入染尘2 h)、4 h组(每天吸入染尘4 h)、8 h组(每天吸入染尘8 h)和对照组(不染尘),共染尘20 d。染尘期间,密切观察各组小鼠的一般情况。染尘结束后,立即称取小鼠的体重和肺重,计算肺脏系数。抽提肺组织总RNA,实时荧光定量PCR(RT-q PCR)法检测各组NF-κB p65 mRNA和TGFβ1 mRNA的表达。结果各暴露时间组小鼠出现早期肺脏损伤的表现:与对照组比较,4和8 h组小鼠的体重降低,肺重上升,差异均有统计学意义(P<0.05);各暴露时间组小鼠的肺脏系数均高于对照组,差异有统计学意义(P<0.05)。对照组和3个暴露时间组NF-κB p65 mRNA相对含量分别为(1.03±0.68)、(1.32±0.84)、(2.17±0.73)、(3.09±2.63);4组间的差异有统计学意义(F=6.68,P=0.00),与对照组比较,4和8 h组NF-κB p65 mRNA表达升高,差异有统计学意义(P<0.05)。对照组及各暴露时间组TGF-β1 mRNA相对含量分别(1.10±0.83)、(4.69±8.97)、(5.66±3.85)、(12.08±13.94),4组间的差异有统计学意义(F=4.24,P=0.01),与对照组相比,各暴露时间组TGF-β1 mRNA的表达均升高,差异有统计学意义(P<0.05)。NF-κB p65 mRNA和TGFβ1 mRNA之间呈正相关关系(r=0.96,P=0.04)。结论矽尘吸入可使小鼠肺组织NF-κB p65 mRNA和TGFβ1 mRNA的表达均升高,两者之间呈正相关。 Objective To investigate the effects of silica dust on the expression of NF-B p65 mRNA and TGFβ1 mRNA in mice. Methods Forty Kunming mice were randomly divided into 4 groups with 10 in each group. There were 2h group ( exposed to dust for 2 h every day) , 4h group (exposed to dust for 4 h every day) ,8h group (exposed to dust for 8 h every day) and a control group (no exposed to dust). The general situation of the mice was observed during the exposal stage. After exposure to dust for 20 days, the weights of body and lung were measured to calculate the lung coefficient of mice. And the total RNA was extracted from the lung tissue to detect the expressions of NF-KB p65 mRNA and TGFβ1 mRNA by Reahime fluorescence quantitative PCR (RT-qPCR) technology. Results The performance of early lung injury appeared in exposal time groups. Compared with the control group, the body weight of 4 and 8 h groups decreased, but the lung weight of 4 and 8 h groups increased, there both were statistically difference (P 〈 0. 05 ). The lung coefficient in each exposal time group was higher than the control group, there was statistical difference ( P 〈 0.05 ). The relative contents of NF-κB p65 mRNA of the control group and 3 exposal time groups were ( 1.03 ±0. 68) , ( l. 32 ±0. 84) , (2. 17 ±0. 73) , (3.09 ±2. 63). There were significant differences among them (F = 6. 68, P = 0. 00). Compared with the control group, the relative contents of NF-KB p65 in 4 and 8 h group significantly increased ( P 〈 0. 05 ). The relative contents of TGF[31 mRNA in the control group and 3 exposal time groups were ( 1.10 ±0. 83 ), (4. 69 ± 8.97 ), (5.66 ± 3.85 ) , ( 12.08 ± 13.94) respectively. There were significant differences among the 4 groups ( F = 4. 24, P = 0. 0l ). Compared with the control group, the relative contents of all exposal time groups were significant higher (P 〈 0.05 ). The correlation between NF-κB p65 mRNA and TGFβ1 mRNA was r = 0. 956, P = 0. 04. Conclusion The expression of NF-κB p65 mRNA and TGFβ1 mRNA in lung tissue of mice exposed to silica dust were all increased. There was a positive relationship between the two indicators.
