小桐子雌雄花性别分化过程中潜在控制基因挖掘 被引量:2

摘要 指出了小桐子雌雄比例失调,极大地降低了小桐子的产量。从小桐子花入手,通过分子手段来增加雌花比例,进而提高小桐子产量。通过SSH和RNA-seq相结合的方法,共找到候选基因基因3个。并利用premer premier5.0对3个基因设计引物,以EF1α为内参,进行qRT-PCR。结果表明:JCGZ_23177在雌花中高效表达,JCGZ_06508和JCGZ_17929在雄花中高效表达,通过NCBI数据库分析,3个基因分别为S-腺苷甲硫氨酸合酶1(S-adenosylmethionine synthase 1)、肽甲硫氨酸亚砜还原酶A5(peptide methionine sulfoxide reductase A5)、脂氧合酶6(lipoxygenase 6)。 The proportion of male and female of Jatropha curcas is greatly reduced,which greatly reduces the yield of Jatropha curcas.In this paper,the proportion of female flowers was increased by molecular means,and then the yield of Jatropha curcas was increased.Through the combination of SSH and RNA-seq method,three candidate genes were found.This paper used primer premier 5.0to design primers for 3genes to examine qRT-PCR with EF1αas the internal reference.The results showed that the expression of JCGZ_23177was highly in female flowers as well as JCGZ_06508and JCGZ_17929in male flowers.Through the NCBI database,the three genes were S-adenosylmethionine synthase 1,peptide methionine sulfoxide reductase A5 and lipoxygenase 6.
