摘要
Toll样受体4(TLR4)-髓样分化因子88(MyD88)-核转录因子κB(NF-κB)信号通路是疾病理变反应中的关键通路,广泛存在于各组织细胞中,是介导炎性因子在细胞内与细胞之间相互表达的重要信号通路之一。研究发现TLR4-MyD88-NF-κB信号通路在肝免疫异常、炎症反应触发的肝损伤、肝星状细胞的活化及肝纤维化恶化与肝癌的发展中均有重要调节作用。该信号通路在"肝炎-肝纤维化-肝癌轴"(IFC轴)的动态变化与肝癌的转移中可能是一个关键的调节点。本文就TLR4-MyD88-NF-κB信号通路参与IFC轴疾病病理机制的最新研究进展作一综述,为以后相关研究提供参考。
As a critical pathway in the reaction of hepatic disease, Toll-like receptor 4 (TLR4)-myeloid differentiation factor 88 (MyD88)-nuclear transcription factor (NF-KB) signalling pathway, which widely exists in various tissues and cells, is one of the most important signalling pathways that mediate the expression of inflammatory factor between the intracellular and intercellular. Study found that TLR4-MyD88-NF-KB signalling pathway plays an important role in the regulation of liver immune abnormalities, inflamma- tory response triggered by the liver damage, activation of hepatic stellate cells and liver fibrosis deterioration and the development of cancer of the liver. The signalling pathway may be an important regulatory point in the dynamic changes of the "hepatic inflammation- fibrosis-cancer axis" (IFC axis) and liver cancer metastasis. In this paper, we review the recent advances in the pathological mecha- nism of TLR4-MyD88-NF-KB signalling pathway participating in 1FC axis disease, so as to provide reference for related research in the future.
