检测T细胞免疫球蛋白及黏蛋白分子3(T cell immunoglobulin and mucin domain containing molecules 3,TIM-3)在胃癌患者固有免疫细胞(γδT细胞与NK细胞)中的表达率,探讨其与临床病理特征及术后中位无进展生存期(progress free survival,PFS)的关系。收集临床病理资料齐全的胃癌患者26例,经流式细胞技术检测TIM-3在胃癌组织、癌旁组织及外周血γδT细胞、NK细胞中的表达率,并分析其与临床病理因素及中位PFS的相关性。结果发现,TIM-3在胃癌组织和癌旁组织中γδT细胞上均表达,而2类组织中γδT细胞上TIM-3的表达率差异无统计学意义(P=0.074),但均显著高于其在外周血γδT细胞上的表达水平(P<0.001;P<0.001)。TIM-3在外周血NK细胞中表达率较高,且TIM-3在外周血中NK细胞表达率显著高于胃癌组织及癌旁组织(P<0.001;P<0.001),而癌组织与癌旁组织之间差异无统计学意义(P=0.097)。NK细胞中TIM-3的表达率分别与肿瘤浸润深度和TNM分期相关(P<0.01),γδT细胞中TIM-3表达率与TNM分期有关(P<0.01)。γδT细胞及NK细胞中TIM-3表达率较低的胃癌患者比TIM-3表达率较高的胃癌患者的中位PFS更长(P=0.014;P=0.028),提示TIM-3在胃癌的进展过程中起到重要作用。
To investigate the expression of T cell immunoglobulin and mucin molecule containing molecules 3 (TIM 3) inγδT cells and NK cells of gastric cancer patients, and explore its correlation with clinical pathologic features and postoperative progression free survival(PFS), we collected 26 cases of gastric cancer patients who had complete clinical and pathological data. The expression of TIM-3 on γδT cells and NK cells in the gastric cancer tissues, the adjacent tissues and the peripheral blood was detected by flow cytometry. The expression of TIM-3 on γδT cells in gastric cancer tissues and adjacent tissues was signifi cantly higher than that in PBMC(P 〈 0. 001 ; P 〈0. 001), but there was no significant difference between the gastric cancer tissues and adjacent tissues(P=0. 074). The expression of TIM-3 in peripheral blood NK cells was higher than that in gastric cancer tissues and adjacent tissues(P〈 0. 001 ; P 〈 0. 001), but there was no statistically significant difference between the last two groups(P=0. 097). The expression of TIM-3 on NK cells in gastric cancer tissues was correlated with tumor depth and TNM stage(P〈 0.01), whereas the expression of TIM-3 on γδT cells in gastric cancer tissues was correlated with TNM stage(P〈 0.01). Patients with lower TIM-3 expression on γδT and NK cells in gastric cancer tissues had longer median PFS than those who with high TIM 3 expression levels(P= 0. 014;P= 0. 028), suggesting that TIM-3 plays an important role in the progression of gastric cancer.
