六味地黄丸在肺癌发生早期对TNF-α和PCNA的影响 被引量:5

摘要 目的:研究六味地黄丸在诱发性肺癌发生早期阶段,对小鼠肺组织中肿瘤坏死因子α和增殖细胞核抗原的影响。方法:腹腔注射乌拉坦制备小鼠诱发性肺癌模型,注射后立即以高、中、低剂量的六味地黄丸连续灌胃给药,正常对照组和肿瘤对照组则给以蒸馏水灌胃,30天后处死小鼠取肺组织,ELISA法检测小鼠肺组织匀浆液中TNF-α的含量,免疫组化法检测肺组织中PCNA的含量。结果:在诱发性肺癌发生早期阶段,与肿瘤对照组比较,中、低剂量六味地黄丸组小鼠肺组织中TNF-α和PCNA的含量无统计学意义上的差别,但高剂量六味地黄丸组小鼠肺组织中TNF-α和PCNA的含量均明显下降。结论:在肺癌发生早期阶段,中、低剂量的六味地黄丸对小鼠肺组织中TNF-α和PCNA的表达无显著影响,高剂量的六味地黄丸早期干预可以降低炎症因子TNF-α的释放,下调PCNA的表达。 Objective: To observe the effect of early invention with high, medium and low dose Liuwei Dihuang Pill(LWDH)on the lev-els of TNF - α and PCNA in the lung tissues at the early stage of induced lung oncogenesis. Method: For induction of chemical multi-stage lung carcinogenesis,mice received twice intraperitoneal( i. p. ) injections of urethane,then high, medium and low dose LWDH were continuously intragastric administered twice one day after urethane start, whereas normal control mice and tumorous control mice received distilled water. The mice were sacrificed after 30 days, and the levels of Tumor Necrosis Factor alpha (TNF - α) in the ho-mogenate of lung tissues were tested with enzyme linked immunosorbent assay. The expression of PCNA were examined with immuno- histochemical technique. Result: At the early stage of induced lung carcinogenesis, compared with tumorous control group, there was no significant difference in the content of TNF - a and PCNA in the groups of medium and low dose LWDH, but high dose LWDH can obviously decrease the content of TNF - α and PCNA in the lung tissues. Conclusion: At the early stage of induced lung cancer, there was no significant effect on the expression of TNF -α and PCNA in the lung tissue of mice with medium and low dose of LWDH, and high dose of LWDH can suppress the release of inflammatory factors TNF -α and reducing the expression of PCNA.
