摘要 目的:研究安石榴苷对慢性支气管炎大鼠抗氧化能力的影响及其作用机制。方法:采用混合烟雾吸入法建立烟熏致大鼠慢性支气管炎模型,将大鼠随机分为:空白对照组、模型组、不同浓度安石榴苷(10、20及40mg/kg)处理组。处理结束后测定各组大鼠肺组织及血清中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性以及丙二醛(MDA)的含量。ELISA法检测各组大鼠血清中TNF-α、IL-8,IL-1β、IL-6的含量。Westernblot法检测PPARγ、PGC-1α、Nrf2、γ-GCS的表达。结果:模型组大鼠肺组织及血清中的SOD、CAT、GSHPx活性均显著低于对照组,MDA含量显著高于对照组。与模型组相比较,20、40mg/kg安石榴苷可显著增加大鼠肺组织及血清中的SOD、CAT、GSH-Px的活性,抑制MDA的产生。ELISA结果表明20、40mg/kg安石榴苷可显著抑制TNF-α、IL-8、IL-1β、IL-6的产生。Western blot结果表明安石榴苷可显著促进PPARγ、PGC-1α、Nrf2、γ-GCS的表达。结论:安石榴苷通过活化PPARγ/PGC-1α和Nrf2/γ-GCS信号通路,进而提高慢性支气管炎大鼠的抗氧化能力,缓解炎性反应。 Objective: To investigate the effect of Puniealagin (PUN) on the activity of antioxidant of chronic bronchitis rats and the underlying mechanism. Methods: Chronic bronchitis rat model was established by passively exposed to smoke mixture. The rats were randomly divided into following groups : controlled group, model group, 10 mg/kg of PUN group, 20 mg/kg of PUN group, 40 mg/kg of PUN group. After treatment, the activity of SOD, CAT and GSH-Px, and the level of MDA in lung tissues and serum were measured. The content of TNF-α, IL-8, IL-1β and IL-6 in serum were measured by ELISA. The expression of PPARγ, PGC-1α, Nrt2 and γ-GCS was measured by Western blot. Results: The activity of SOD, CAT and GSH-Px were signifi-cantly decreased, and the content of MDA was increased in lung tissues and serum of model rats compared with control group. Whereas after the treatment of PUN, the activity of SOD, CAT and GSH-Px were significantly increased, and MDA was decreased compared with model group. The results of ELISA showed that PUN significantly decreased the content of TNF-α, IL-8, IL-113 and IL-6 in serum of model rats. The results of Western blot showed that PUN increased the expression of PPART, PGC-1α, Nrf2 and γ-GCS. Conclusion: PUN can increase the activity of antioxidant and decrease the inflammatory response through acti- vating the PPARγ/PGC-1α and Nrf2/γ-GCS pathway in chronic bronchitis rats.
