妊娠相关蛋白A的多态性与冠心病的相关性研究

Study on the correlation between PAPP-A polymorphism and coronary heart disease

目的 探讨冠心病患者的妊娠相关蛋白A（PAPP-A）基因多态性与冠心病之间的关系.方法 以2014年1月至2015年2月到我院就诊的冠心病患者共100例作为研究对象,为试验组;选取同期到我院进行体检的健康人共100位作为对照组.首先对收集的受试者血液样本进行DNA提取,然后使用PCR方式进行基因扩增,PCR的产物则使用ECOR V内切酶进行酶切,使用PAPP-A扩增酶切产物,使用琼脂糖（2％）进行凝胶电泳以鉴定PAPP-A的基因型,观察PAPP-A的基因多态性.结果 PAPP-A基因型共有3种,分别是AA型、CC型两种纯合子和AC型一种杂合子.AA型酶切后表现为77 bp一个条带;CC型酶切后表现为107 bp和77 bp两个条带;AC型酶切后则表现为107bp、77bp和30 bp三个条带.试验组CC型基因型明显多于对照组,C型等位基因的频率高于对照组（P＜0.05）,差异具有统计学意义.结论 在冠心病患者体内PAPP-A的基因型发生了明显的变化,主要表现为CC型的基因型增多,C等位基因的频率提高,因此PAPP-A可能与冠心病的发病具有一定的关系,为临床诊断和治疗提供了一定的依据.

Objective To investigate the polymorphism of PAPP-A gene in patients with coronary heart disease,and to explore the relationship between coronary heart disease and coronary heart disease.Methods A total of 100 patients with coronary heart disease in our hospital from February to January 2014 in our hospital were selected as the research subjects of this study,the experimental group was-2015.Select the same to our hospital for health examination of healthy people a total of 100 cases as the control group,first to collect blood samples for DNA extraction,and then use PCR gene amplification,the PCR products using ECOR V enzyme digestion and use PAPP-A amplification enzyme digestion products using agarose （2%） gel electrophoresis to identify PAPP-A gene type was observed PAPP-A gene polymorphism.Results PAPP-A gene type of a total of three,respectively was AA and CC homozygotes and AC type a heterozygous and AA type enzyme digestion performance 77 bp 1 a belt,CC type enzyme digestion is performance for a 107 bp,77 bp two bands,AC enzyme digestion is a 107 bp and 77 bp and 30 bp with three.The experimental group CC genotype was significantly higher than the control group,the experimental group,the frequency of C allele was higher than the control group （P〈0.05）,the difference was statistically significant.Conclusion PAPP-A in patients with coronary heart disease （CHD） genotypes in occurred obvious change,mainly as an increase in the CC genotype,increase the frequency of C allele,so PAPP-A may has some relation with the incidence of CHD,provides a certain basis for clinical diagnosis and treatment.
