脑胶质瘤患者脑脊液中HOXA13、Plexin-B2的定量分析及生物学意义研究 被引量:2

摘要 目的探讨同源基因A-13(homeotic genes-A13,HOXA13)和神经丛素-B2(Plexin-B2)在脑胶质瘤中的表达情况及生物学意义。方法收集2014年5月~2016年2月间我院诊断的脑胶质瘤患者60例,使用qRT-PCR技术分别检测实验组(脑胶质瘤患者)和对照组(健康受试者)血清中S100、HOXA13和Plexin-B2的表达水平,并分析HOXA13和Plexin-B2在不同级别脑胶质瘤患者中的表达情况,分析其与肿瘤恶性程度的关系。结果与对照组对比,实验组的HOXA13和Plexin-B2在脑脊液及血浆中表达水平显著升高[血液中:HOXA13,(1.21±0.14)vs 0;Plexin-B2:(1.57±0.33)vs 0;脑脊液中:Plexin-B2:(4.87±1.13)vs 0;Plexin-B2:(5.88±0.21)vs 0],肿瘤体积越大、分化程度越低,血液中HOXA13和Plexin-B2表达水平越高;脑脊液中HOXA13和Plexin-B2蛋白水平明显高于血浆中。结论脑胶质瘤患者中HOXA13和Plexin-B2表达上调,且与肿瘤恶性程度有显著关系,脑脊液中HOXA13和Plexin-B2表达显著升高,可用作脑胶质瘤恶性程度的标志物。 Objective To investigate the expression and biological significance of gomeotic genes-A13(HOXA13) and neuronal-B2 (Plexin-B2) in gliomas. Methods 60 patients diagnosed with glioma were diagnosed in our hospital from May 2014 to February 2016 were chosen. The serum expression levels of S100, HOXA13 and Plexin-B2 in the experimental group (glioma patients) and the control group (health volunteers) were measured by qRT-PCR. And the expression of HOXA13 and Plexin-B2 in different stages of glioma patients was analyzed, the relationship with the degree of tumor ma[ignancy was analyzed. Results Compared with those of the control group, the expression levels of HOXA13 and Plexin-B2 in cerebrospinal fluid and plasma were significantly higher [blood: HOXA,13,(1.21±0.14) vs O; Plexin-B2, (1.57±0.33) vs 0; cerebrospinal fluid: Plexin-B2, (4.87±1.13) vs O; Plexin-B2, (5.88±0.21) vs 0]. The larger the tumor volume, the lower the degree of differentiation and the higher the expression level of HOXAI3 and Plexin-B2 in the blood. The levels of HOXA13 and PIexin-B2 protein in cerebrospinal fluid were significantly higher than those in plasma. Conclusion The expression of HOXA13 and Plexin-B2 in brain glioma patients is up-regulated and is significantly correlated with the degree of malignancy. The expression of HOXA13 and Plexin-B2 in cere- brospinal fluid is significantly increased and can be used as a marker of malignant degree of glioma.
