摘要
目的探讨胃癌干细胞CSC-G在胃癌侵袭和转移中的作用。方法体外培养胃癌干细胞CSC-G和普通胃癌细胞SGC7901采用Western blot法检测两种胃癌细胞中干细胞标志物的表达情况,对比两种胃癌细胞的侵袭性检测结果和球形克隆形成情况。结果胃癌干细胞CSC-G的干细胞标志物SOX2、OCT4的表达水平高于胃癌细胞SGC7901(P<0.05)。球形克隆检测结果显示,胃癌干细胞CSC-G的瘤球数量为(65.49±10.22)个,多于胃癌细胞SGC7901的(7.36±1.14)个(P<0.05)。侵袭能力检测结果显示,胃癌干细胞CSC-G穿过基底膜的数量为(168.71±32.50)个,多于胃癌细胞SGC7901的(92.44±13.06)个(P<0.05)。结论胃癌干细胞CSC-G在胃癌侵袭和转移过程中发挥着促进作用。
Objective To investigate the role of CSC-G in gastric cancer stem cell invasion and metastasis. Methods The expression of stem cell markers in gastric cancer stem cells (CSC-G) and SGC7901 cells weredetected by Western blot. The invasive test results and the formation of spherical clones were compared between the two gastric cancer cells. Results The expression levels of SOX2 and OCT4 in gastric cancer stem cells were significantly higher than those in gastric cancer cells SGC7901 (P 〈 0.05). The number of tumor cells in gastric cancer stem cells was (65.49±10.22), which was more than that of gastric cancer cells SGC7901 (7.36±1.14) (P 〈 0.05). The number of CSC-G through gastric basement cells was (168.71±32.50); which was higher than that of gastric cancer cell SGC7901 (92.44±13.06) (P 〈 0.05). Conclusion CSC-G plays an important role in the invasion and metastasis of gastric cancer.
