铁调素基因敲除后对铁蓄积小鼠骨代谢指标的变化 被引量:3

Effects of hepcidin gene knock out on bone turnovers in mice with iron accumulation

摘要 目的观察铁调素基因敲除后铁蓄积小鼠骨代谢指标变化。方法C57背景小鼠分为野生型(WT)及铁调素基因敲除小鼠(KO),每组各5只雄性,检测小鼠血清铁蛋白、血清成骨指标碱性磷酸酶(ALP)、血清破骨指标抗酒石酸酸性磷酸酶(TRAP-Sb)、股骨远端micro—CT扫描、骨组织相关基因。结果血清铁蛋白WT组(4.88±0.74)μg/L,KO组血清铁蛋白(17.62±0.84)μg/L,KO组显著升高(P=0.013);WT组血清ALP(9.42±0.74)ng/ml,KO组血清ALP(5.41±0.63)ng/ml,KO组显著降低(P=0.038);WT组血清TRAP-Sb(72.36±1.85)ng/ml,KO组血清TRAP-Sb(132.64±2.16)ng/ml,KO组显著升高(P=0.018);micro—CT三维重建显示KO组骨量丢失;骨组织实时定量聚合酶链反应(Real—timePCR)结果显示KO组核心结合蛋白因子2(Runx2)基因表达显著下降,TRAP-5b基因表达显著上升。结论铁调素基因敲除后铁蓄积小鼠模型骨量丢失,成骨指标受抑制,破骨指标活跃。 Objective To explore the iron accumulation after hecidin gene knock out mice bone turnovers change research. Methods C57 background mice were divided into wild type mice (WT) and hepcidin gene knockout mice (KO), each group has 5 male mice, all mice test serum ferritin and serum alkaline phosphatase (ALP) osteogenesis index, serum index of osteoclasts tartrate -resistant acid phosphatase 5b (TRAP -5b), distal femur micro -CT scan, bone tissue related genes expression. Results Serum ferritin WT group (4. 88 ±0. 74) μg/L, KO group serum ferritin (17. 62 ±0. 84) μg/L, KO group increased significantly (P =0. 013) ; WT group serum ALP (9.42 ±0. 74) ng/ml, KO group serum ALP (5.41 ± 0.63) ng/ml, KO group significantly reduced (P= 0.038) ; Serum TRAP - 5b WT group (72. 36 ± 1.85) ng/ml, KO group serum TRAP -5 b ( 132. 64 ±2. 16) ng/ml, KO group increased sig- nificantly (P =0. 018) ; Micro - CT three dimensional reconstruction show KO group of bone loss; Bone tissue real- time quantitative polymerase chain reaction (Real- time PCR ) results showed KO group Runx2 gene expression decreased significantly, the TRAP- 5b gene expression increased. Conclusion uppressed, osteoclasts indicators were actived.
