摘要
目的探讨长链非编码RNASH3PXD2A-AS1在胃癌术后肝转移诊断中预测价值。方法收集37例胃癌及其配对正常胃黏膜组织,采用荧光实时定量聚合酶链反应(PCR)方法检测组织中SH3PXD2A—AS1表达水平,分析其表达与患者性别、年龄、肿瘤部位、肿瘤直径、组织分化程度、肿瘤浸润深度、淋巴结转移、肝转移数目及肝转移发生时间等临床病理因素的关系。并作统计学分析处理。结果在37例胃癌组织中SH3PXD2A—AS1阳性率为89.19%(34/37),而正常胃组织SH3PXD2A—ASI阳性率为5.41%(2/37)。两者差异有统计学意义(x^2=55.392,P=0.000)。SH3PXD2A—AS1表达量与患者性别(x^2=1.718,P=0.190)、年龄(x^2=0.560,P=0.756)、肿瘤位置(x^2=2.839,P=0.242)、组织分化程度(x^2=0.924,P=0.336)均无明显相关,但与原发灶直径(x^2=4.788,P=0.029)、肿瘤浸润深度(x^2=8.815,P=0.012)、淋巴结转移(x^2=6.801,P=0.009)、肝转移数目(x^2=10.157,P=0.001)、肝转移发生时间(x^2=6.903,P=0.032)均显著相关。结论SH3PXD2A-AS1在胃癌术后肝转移诊断中具有预测价值。
Objective To investigate the predictive value of long chain non coding RNA SH3PXD2A - AS1 in the diagnosis of postoperative liver metastasis of gastric cancer. Methods Human gastric cancer and matched normal gastric tissues were collected, and SH3PXD2A - AS1 was detected by fluorescence quantitative realtime polymerase chain reaction (PCR). The relationship between SH3PXD2A- AS1 expression and patient's sex, age, tumor location, tumor diameter, histological differ- entiation, depth of tumor invasion, lymph node metastasis, the number of liver metastases and the time of liver metastasis distant metastasis was analyzed, respectively. Results The positive rate of SH3PXD2A - AS1 in 37 cases of gastric cancer was 91.89% (34/37) , and that in normal gastric tissues was 5.40% (2/37). The difference was very significant (x^2 = 55. 392, P = 0.000). SH3PXD2A - AS1 expression was not significantly correlated with the age (x^2 =0. 560,P =0. 756) and sex (x^2 = 1. 718,P =0. 190), tumor location (x^2 = 2. 839, P = 0. 242 ), histological differentiation (x^2 = 0. 924, P = 0. 336 ), but with the pri- mary tumor diameter (x^2 = 4. 788,P = 0. 029), depth of tumor invasion (x^2 = 8. 815, P = 0. 012), lymph node metastasis (x^2 = 6. 801,P = 0. 009), the number of liver metastases (x^2 = 10. 157, P = 0. 001 ), and liver metastasis time (x^2 = 6. 903, P = 0. 032). Conclusion SH3PXD2A - AS1 has important predictive value in the diagnosis of postoperative liver metastasis of gastric cancer.
