大蒜素对皮肤基底细胞癌细胞放射治疗敏感性的影响及其与自噬水平的关系

摘要 目的探讨大蒜素(Allicin)对皮肤基底细胞癌细胞放射治疗敏感性的影响及其与自噬水平的关系。方法采用MTT法检测Allicin对A431细胞的生长抑制作用,记录半数抑制浓度(IC50);将细胞分为4组:照射组(IR组)、Allicin组、IR+Allicin组以及对照组。细胞自噬水平的检测采用流式细胞仪。结果MTT检测结果显示,A431细胞经Allicin处理24,48,72h后,其活性受到显著的抑制,抑制程度受到时间及浓度的影响。同一浓度下,不同时间比较以及同一时间内不同浓度的比较,A431细胞活性的差异均具有统计学意义(P均<0.05)。Allicin作用于A431细胞24,48,72h的IC50分别为34.14,17.78,6.72mmol/L,差异均具有统计学意义(P均<0.05)。IR+Allicin组的SF_2,D_0,D_q分别为:75.81,6.46,1.34;IR组的SF_2,D_0,D_q分别为96.21,8.60,2.85。在各照射剂量点A431细胞的SF均显著低于IR组。与IR组比较,IR+Allicin组的肩区逐渐缩小,说明细胞的损伤修复能力呈现降低趋势。Allicin组和IR+Allicin组的A431细胞自噬水平均高于对照组,差异有统计学意义(P均<0.05)。结论 Allicin能上调人皮肤基底细胞癌A431细胞的自噬水平,进而增加A431细胞的放疗敏感性。 Objective To investigate the effect of Allicin on the radiosensitivity of human skin basal cell carcinoma cells and its relationship with autophagy level. Methods MTT was used to detect the inhibitory effect of Allicin on A431 cells. IC50 was recorded. Cells were divided into four groups: irradiated group (IR), Allicin group, IR + Allicin group and control group. Cell autophagy was detected by flow cytometry. Results The results of MTF showed that A431 cells were significantly inhibited by Allicin for 24, 48 and 72h, and the in- hibition was affected by time and concentration. Comparison of the same concentration at different times, the comparison of different concentrations at the same time, the activity of A431 cells was significantly different (all P 〈0. 05). The IC50 of Allicin on A431 cells at 24, 48 and 72h was 34. 14, 17. 78, 6. 72mmoL/L (all P 〈0. 05), all the differences were statistically significant. The SF2, Do and Dq of IR + Allicin group were 75.81, 6. 46 and i. 34, respectively. The SF2 ,D0and Dq of IR group were 96. 21, 8.60 and 2. 85 respectively. The SF of A431 cells was significantly lower than that of IR group at each dose. Com- pared with the IR group, the shoulder area of the IR + A11icin group decreased gradually, indicating that the injury repair capacity of the IR + Allicin group decreased. The autophagy level of A431 cells in Allicin group and IR + Allicin group was higher than that in control group ( all P 〈 0. 05 ) , the differences were statistically significant. Conclusion Allicin can up-regulate the autophagy of human skin A431 cells and in- crease the radiotherapy sensitivity of A431 cells.
