摘要
为探讨食淀粉乳杆菌抗猪轮状病毒的可能机制,选择3日龄清洁级中国昆明鼠乳鼠为实验动物,每天灌喂食淀粉乳杆菌,在饲喂后第4天接种轮状病毒,应用ELISA定量检测猪轮状病毒的主要受体之一神经节苷酯(Ganglioside)GM3,并采用实时荧光定量PCR技术测定各样本中葡萄糖神经酰胺糖基转移酶(UGCG)、ST3-半乳糖苷α-2,3唾液酸转移酶5(St3gal5)的mRNA相对含量。结果显示,饲喂后第5、9及11天,食淀粉组神经节苷酯GM3含量显著高于正常组及攻毒组(P<0.05),第5、7、9及11天时食淀粉组Mus St3gal5 mRNA相对表达量均显著低于正常组(P<0.05),第7、9及11天时食淀粉组Mus St3gal5 mRNA相对表达量显著低于攻毒组(P<0.05),第7、9及11天时食淀粉组Mus UGCG mRNA相对表达量均显著低于正常组及攻毒组(P<0.05)。结果表明,乳鼠感染轮状病毒后,食淀粉乳杆菌能够显著促进小鼠空肠中神经节苷酯GM3的合成,并显著抑制小鼠空肠中Mus St3gal5 mRNA及Mus UGCG mRNA的表达。本研究为进一步研究食淀粉乳杆菌抗轮状病毒机理提供了帮助。
To investigate the possible mechanisms of Lactobacillus amylovorus against porcine ro- tavirus(PRV),3-day-old clean grade Kunming mice were fed with L. amylovorus for 4 days,and the ro- tavirus was inoculated on the 4th day of the experiment. One of the major receptors of porcine rotavirus (Ganglioside) GM3 was quantitatively detected by ELISA,and the mRNA relative contents of UDP-glucose ceramide glucosyltransferase(UGCG) and ST3 beta-galactoside alpha-2,3-sialyltransferase 5(St3gal5) in each sample were determined by real-time fluorescence quantitative PCR. The results showed that the content of GM3 in the L. amylovorus group was significantly higher than that in the normal group and the virus group on day 5,9 and II(P〈0.05) ,and the relative expression of MusSt3gal5 mRNA in the L. amylo- vorus group was significantly lower than that in the normal group onday5,7,9 and II(P〈0. 05). The relaive expression of MusSt3galSmRNA in the starch group was significantly lower than that in the virus group on day 7,9 and If(P〈0. 05).The relative expression of MusUGCG mRNA in the L. amylovorus group was signifi-cantly lower than that in the normal group and the virus group on day 7,9 and ll(P〈0. 05). Theresults showed that L amylovoruscould promote the synthesis of gangliosideGM3 in the jejunum and inhibit the expres- sion of MusSt3gal5 and MusUGCG mRNA in the jejunum in suckling mice after infection with rotavirus. The study offers help to further study the effects of L. amylovorus against rotavirus mechanisms.
