禽腺病毒血清4型的分离鉴定及遗传演化分析 被引量:15

摘要 本研究从辽宁发病鸡群采集的病料经SPF鸡胚接种传代分离病毒,并其进行了PCR和测序分析鉴定。结果,从病料中经SPF鸡胚接种传代分离出一株禽腺病毒,经PCR和测序分析鉴定确定该分离株为禽腺病毒C种、血清4型,命名为LN160130;通过对52 k和hexon基因同源性比对,LN160130与我国近年报道的禽腺病毒血清4型同源性最高,52 k和hexon基因的同源性均为100%;与国外流行的血清4型毒株相比,LN160130的52k基因同源性99.3%~100%,hexon基因同源性98.7%~99.8%。遗传演化分析结果表明,LN160130分离株与我国禽腺病毒血清4型流行毒株在同一分支内,而与国外禽腺病毒血清4型毒株不在同一分支,存在遗传差异,说明中国流行的血清4型禽腺病毒有自身地域特点。 In this study,a fowl adenovirus strain was isolated from samples of chicken flocks in Liaoning Province by using specific pathogen-free chicken embryo, lt was identified by PCR and sequencing,and designated as LN160130,serotype 4 in species C,genus aviadenovirus,family aden- oviridae. Multiple alignment based on the 52k and hexon genes indicated that,the isolate of LN160130 showed the highest identity of 100% with the Chinese prevalent serotype 4 strains, ln comparison with the foreign strains,the isolate LNI60130 showed from99.3% to 100% homology for 52k gene and from 98.7% to 99.8% identity for hexon gene. Evolutionary analysis showed that iso- late LN160130 formed a distinct branch with the chinese strains beside the foreign strains. The chinese prevalent serotype 4 possessed the unique geographic features different from the foreign strains.
