摘要
利用新一代高通量测序手段对大针茅受到羊啃食后2h的实验组与未啃食的对照组叶片进行Illumina平台的转录组测序(RNA sequences),共得到147561个序列读取片段(reads),组装成64738条单基因簇(Unigenes),Unigenes平均长度为1367.21bp,N50为2017bp。随机挑选11个基因利用实时荧光定量PCR对转录组数据库进行了验证。基于转录组数据分析淀粉和蔗糖代谢通路中关键调控基因表达差异,结果表明:随机挑选的11个基因的表达差异变化与转录组数据上下调趋势一致,在淀粉与蔗糖代谢通路中相关基因的表达差异变化利于淀粉、蔗糖的积累。大针茅在羊啃食后淀粉与蔗糖含量的增加有利于结实,对后代的传播意义重大。
Using the new means of high-throughput sequencing the transcrlptome sequencing (RNA sequences) of leaf of Stipa grandis grazed by sheep for two hours and no-grazed by sheep (CK) was done on the plate of Illumina, 147561 reads were got, assembled into 64738 single genes, the average length of single genes was 1367.21bp, N50 was 2017bp. Based on the transcriptional data, the key regulatory genes in starch and sucrose metabolism were up-regulated. 11 genes were randomly selected to be verified by real-time fluorescence quantitative PCR. The results showed that the random selected 11 genes expression was consistent with transcriptome data in downward trend. The increase of content of starch and sucrose in Stipa grandis after grazed by sheep was conducive to bear fruit, which is of great significance to the spread of the offspring.
