HMG对小鼠卵母细胞外成熟的影响

摘要 目的探讨人绝经期促性腺激素(HMG)对卵母细胞体外成熟(IVM)的影响。方法选取8~12周龄雌鼠,腹腔注射HMG,48h后处死取卵巢,穿刺卵巢表面获得卵母细胞。将所得卵母细胞随机分成二组。对照组添加FSH、HCG各75IU/L,实验组为HMG组FSH、LH各75IU/L。18h后对成熟卵行单精子显微注射,比较各组的受精率、卵裂率、优质胚胎率。结果 (1)二组卵母细胞成熟率、受精率、卵裂率的差异无统计学意义(P>0.05)。(2)HMG组优质胚胎率较高,相比对照组差异有统计学意义(P<0.05)。结论 IVM培养液添加HMG有利于胚胎的发育。 Objective To explore the effect of HMG on mice oocytes matueation in vitro. Methods Immature oocytes were taken from mice stimulated by intraperitoneal injection of HMG after 48 hours, and all the eggs were divided into two groups. The concentration of FSH&HCG in control group was 75IU/L, while the concentration of FSH&LH in experimental group was 75IU/L. The maturate oocytes were implemented by ICSI microinjection technology after 18 hours. The fertilization rates, cleavage rates, high- quality embryo rates were compared among the groups. Results There was no statistical significance between the two groups of oocytes maturation rate, fertilization rate and cleavage rate (P〉0.05). The experimental group had the better high-quality embryos rate than the control group (P〈0.05). Conclusion HMG added into IVM medium is conducive to the development of embryo.
