阿魏酸钠对心肌缺血/再灌注损伤大鼠能量代谢的影响 被引量:9

Effects of Sodium Ferulate on Energy Metabolism in Rats with Myocardial Ischemia/Reperfusion Injury

摘要 目的:探讨阿魏酸钠对心肌缺血/再灌注损伤大鼠能量代谢的影响。方法:将SD大鼠随机分为:阿魏酸钠高剂量组(40 mg·kg-1)、阿魏酸钠低剂量组(20 mg·kg-1)、假手术组、模型组(n=10),通过结扎冠脉法建立心肌缺血/再灌注损伤模型,各组于冠脉结扎前5 min和再灌注前5min各静脉注射给予1/2量的药物;再灌注结束后,采用比色法测定血清肌酸激酶(CK)和乳酸脱氢酶(LDH)活性,采用定磷法测定心肌组织Na+/K+-ATP酶和Ca2+/Mg2+-ATP酶的活力,采用染色法测定心肌梗死面积(MIS)。结果:阿魏酸钠高、低剂量组MIS显著缩小,与模型组比较差异均有统计学意义(P<0.05或P<0.01);与模型组比较,阿魏酸钠高、低剂量组CK活性和LDH活性显著降低(P<0.05或P<0.01);与模型组比较,阿魏酸钠高、低剂量组可显著升高Na+/K+-ATP酶活力(P<0.05或P<0.01),高剂量组可显著升高Ca2+/Mg2+-ATP酶活力(P<0.05)。结论 :阿魏酸钠对心肌缺血/再灌注所致心肌损伤的保护作用可能与改善心肌组织的能量代谢有关。 Objective: To investigate the effects of sodium ferulate on energy metabolism in rats with myocardial ischemia/reperfusion (MI/R) injury. Methods: SD rats were divided randomly into sodium ferulate high (40 mg·kg-1) and low (20 mg·kg-1) dose groups, sham and model groups (n=10). The MI/R injury model was established by coronary artery ligation (30 min) and subsequent reperfusion (2 hours) in rats. Sodium ferulate was administered by intravenous injection at half dose 5 minutes before the ligation and reperfusion, respectively. After the end of reperfusion, the CK and LDH activities in sera were examined by chromatometry, the Na+/K+-ATPase and Ca2 /Mg2 -ATPase activities in brain tissues were detected by phosphorus determination and the myocardial infarction size (MIS) was determined by staining. Results: Sodium ferulate (40 and 20 mg·kg-1) significantly reduced MIS, comparing with those of the model group (P〈0.05 or P〈0.01). The CK and LDH activities in the sodium ferulate high and low dose groups were significantly lower than those of the model group (P〈0.05 or P〈0.01). Sodium ferulate evidently increased the Na+/K+-ATPase activity at high and low dose and significantly increased the Ca2 /Mg2 -ATPase activity at high dose, respectively, comparing with those of the model group (P〈0.05 or P〈0.01). Conclusion: The protective effects of sodium ferulate in MI/R injury appears to be associated with the improvement of energy metabolism.
