microRNA-181b对牛腺垂体细胞FSH分泌影响的研究 被引量:1

摘要 腺垂体细胞分泌的促卵泡激素(FSH)在繁殖的调节中起重要作用。然而,miRNA是否调控FSH的分泌尚不清楚。在本研究中,我们将miR-181b转染到牛腺垂体细胞中,并通过RT-PCR和ELISA对FSHb表达水平和FSH分泌进行了评估。结果表明:与对照组相比,miR-181b mimics下调FSHb的表达水平,降低FSH分泌,并且miR-181b抑制剂能够上调FSHb的表达水平并增加FSH的分泌。研究结果为miRNA调节FSH的分泌提供了理论支撑,并有助于使用miRNA来调节动物繁殖。 Follicle-stimulating hormone (FSH) secreted by adenohypophyseal cells plays an import- ant role in the regulation of reproduction. However, it is unclear whether miRNh regulates the secretion of FSH. In the present study, we transfected the miH-181bmimics and inhibitor into bovine anterioradenohypophysealcells and assessedFSHb expression levels and FSH secretion via quantitative PCR and ELISA. The results showed that miH-181b down-regulated the expression levels of FSHband decreased the secretion of FSH compared with those of the control group, and treatment with miR-181b inhibitor up-regulated the expression levels of FSHband increased the secretion of FSH. Our resultsprovide evidence that miRNAs can regulate the secretion of FSH and may contribute to the use of miRNhs for the regulation of animal reproduction.
