莽草酸抑制大鼠RBL-2H3细胞脱颗粒和组胺释放 被引量:7

摘要 目的研究莽草酸对大鼠RBL-2H3细胞增殖和化合物48/80(C48/80)诱导后细胞脱颗粒情况及其机制。方法MTT法检测(3、10、30)μg/m L的莽草酸处理后,RBL-2H3细胞的增殖情况,甲苯胺蓝染色观察C48/80诱导RBL-2H3细胞脱颗粒的情况,底物法检测(0、12.5、25、50、80、100)μg/m L C48/80处理后,RBL-2H3细胞内β-氨基己糖苷酶的释放,ELISA检测各处理组细胞上清液中组胺的含量。结果 (3、10、30)μg/m L莽草酸对RBL-2H3细胞增殖无明显抑制作用,RBL-2H3细胞脱颗粒与C48/80的剂量有一定量效关系;莽草酸能抑制大鼠RBL-2H3细胞脱颗粒;与阳性对照组相比,莽草酸处理C48/80刺激后的RBL-2H3细胞,其β-氨基己糖苷酶释放率和组胺释放量均显著降低。结论莽草酸可抑制大鼠RBL-2H3细胞脱颗粒,减少组胺释放。 Objective To study the effects of shikimic acid on the proliferation of rat RBL-2H3 cells and the degranulation of the cells induced by C48/80 and its mechanism.Methods MTT assay was performed to measure the proliferation of RBL-2H3 cells treated with 3,10,30 μg/m L shikimic acid.Toluidine blue staining was used to observe the degranulation of RBL-2H3 cells.The release of β-hexosaminidase from RBL-2H3 cells treated with 0,12.5,25,50,80,100 μg/m L C48/80 was determined by substrate assay.ELISA was used to detect the histamine content in the supernatant of each treated group.Results Shikimic acid at 3,10,300 μg/m L had no obvious inhibitory effect on the proliferation of RBL-2H3 cells.There was a dose-effect relationship between the degranulation of RBL-2H3 cells and C48/80 concentration.Shikimic acid inhibited the degranulation of RBL-2H3 cells compared with the positive control group,the β-hexosaminidase release rate and histamine release were significantly reduced in RBL-2H3 cells treated with shikimic acid and C48/80.Conclusion Shikimic acid can inhibit the degranulation of RBL-2H3 cells and reduce histamine release.
