针康法对脑缺血大鼠脑组织微血管内皮超微结构及aFGF、PF4蛋白表达影响 被引量:8

Influence of Acupuncture-Rehabilitation Therapy on Microvascular Endothelial Ultrastructure,Expression of aFGF and PF4 in Cerebral Ischemia Rats

摘要 目的:探讨针康法对脑缺血后脑组织微血管内皮超微结构及酸性成纤维细胞生长因子(aFGF)、血小板第4因子(PF4)蛋白表达影响。方法:180只大鼠随机分为模型组、针刺组、康复组、针康组及假手术组。参照Longa线栓法制备永久的局灶性脑缺血模型。模型组和假手术组不给予任何干预,针刺组应用头穴丛刺针法,康复组应用跑台训练,针康组应用头穴丛刺结合跑台训练。电镜观察各时间点各组大鼠缺血半暗区皮层微血管内皮的超微结构,Western Blot法检测各时间点各组大鼠缺血半暗区皮层aFGF及PF4蛋白表达。结果:微血管内皮超微结构:术后3天,针刺组、康复组及针康组可见整个管腔内充满内皮细胞,管周水肿程度减轻,但针康组的效果较好;术后7天时,针康组内皮细胞增殖明显,微血管内膜平整,管腔无狭窄,而针刺组和康复组微血管内膜较针康组欠平整,微血管管腔狭窄;术后14天时,针康组微血管基膜是完整的,且内皮细胞基本恢复正常,而针刺组及康复组微血管基膜稍显不完整。aFGF及PF4蛋白表达:术后各时间点,较模型组比较,各干预组aFGF蛋白表达明显升高(均P<0.05),PF4蛋白均明显降低(均P<0.05);术后7天、14天,较针刺组、康复组比较,针康组aFGF蛋白表达升高(均P<0.05),PF4蛋白表达降低(均P<0.05)。结论:针康法通过上调缺血脑组织aFGF,下调PF4蛋白表达,降低缺血脑组织血管内皮的损伤。 Objective:To investigate the influence of Acupuncture - Rehabilitation Therapy on microvascular endothelial uhrastructure and expression of acidic fibroblast growth factor (aFGF) and platelet factor - 4 (PF4) in cerebral isehemia rats. Methods:Permanent focal cerebral ischemia was established in rats according to Longa methods. 180 rats were randomly divided into model group( group M), sham group( group C), acupuncture group( group A), rehabilitation group (group R) and acupuncture combined with rehabilitation group (group A -R). No treatment was given to group M and group C. Cluster needling of scalp acupuncture was given to group A. Treadmill training was used for group R. The combination therapy of acupuncture and treadmill training was used for group A - R. Uhrastructure of microvascular endothelial around penumbral cortex area, facilitated factor aFGF and inhibitory factor PF4 were observed and detected by electron microscopy at each time point by Western Blot method. Results:Microvessel endothelia ultrastructure :3 days after modeling, endotheliocytes were filled in lumen and edema around was relieved in group A, group R and group A - R, of which group A - R was better than the other two groups. 7 days after modeling, the proliferation of endotheliocytes was significant, microvascular endangium was smooth and no stricture developed in lumen in group A - R; Unsmooth microvascular endangium was found in the other two groups. 14 days after modeling, microvascular basement membrane was complete and endotheliocytes returned to almost normal in group A -R; Microvascular basement membrane was a bit incomplete both in group A and group R. Expression of revascularization facilitated factor aFGF and inhibitory factor PF4: At various time points, compared with group M, the expression of aFGF was all higher ( P 〈 0.05 ) and the expression of PF4 was all lower ( P 〈 0.05 ) in the three intervened groups. 7 days and 14 days after modeling, the expression of aFGF in group A - R was obviously higher ( P 〈 0.05 ) and the expression of PF4 protein was obviously lower (P 〈 0.05 ) than those in group A and group R. Conclusion : Acupuncture - Rehabilitation Theray can reduce the vascular endothelial cell injury, which may be associated with the up - regulation of aFGF protein expression and down - regulation of PFd protein expression.
