摘要 目的探讨Th17/Th22/Treg细胞及其细胞因子在先兆流产发病中的作用。方法采用流式细胞术检测20例先兆流产组患者和20例正常对照组女性外周静脉血Th17/Th22/Treg细胞频数,采用ELISA双抗体夹心法检测血清中IL-17A、IL-22、IL-23和IL-35细胞因子,RT-PCR检测Th17/Th22/Treg细胞的转录因子(ROR-γt/AHR/Foxp3)的mRNA表达水平。结果先兆流产组的Th17和Th22细胞百分比高于正常对照组;Treg细胞百分比低于正常对照组;Th17/Treg比例失衡,上述差异均有统计学意义(P<0.01)。先兆流产组的IL-17A、IL-22和IL-23表达水平高于正常对照组;IL-35表达水平低于正常对照组;IL-17A/IL-35比例失衡,上述差异均有统计学意义(P<0.01)。先兆流产组外周血中Foxp3的mRNA表达明显低于对照组;AHR和ROR-γt的mRNA表达明显高于正常对照组,上述差异均有统计学意义(P<0.01)。结论 Th17/Th22/Treg细胞失衡可能导致先兆流产发生。 Objective To investigate the role of Th17/Th22/Treg cells and cytokines in the pathogenesis of threatened abor- tion. Methods The peripheral venous blood from 20 cases of threatened abortion and 20 healthy pregnant females selected by physical examination was collected for the measurement of the frequencies of Th17/Th22/Treg cells by flow cytometry(FCM). The serum levels of IL - 17A, IL - 22, IL - 23 and IL - 35 cytokines were measured by the double antibody sandwich ELISA method. The mRNA expression levels of the transcription factors(TFs) of Th17/Th22/Treg cells( ROR - γt/AHR/Foxp3 ) were detected by RT - PCR. Results The percentages of Thl7 and Th22 cells of women with threatened abortion were higher than those of the control group, and the percentage of Treg cells was significantly lower than that of the control group, with obvious Thl7/Treg imbalance, and the differences were statistically significant(P 〈0.01 ). The expression levels of IL- 17A, IL- 22 and IL - 23 in the women with threatened abortion were higher than these of the control group, while the IL - 35 expression level was lower than that of the control group, with obvious IL - 17A/IL -35 imbalance, and the differences were statistically signifi- cant( P 〈 0.01 ). The mRNA expression level of Foxp3 in the peripheral blood of the women with threatened abortion was obvi- ously lower than that of the control group, while the mRNA expression levels of AHR and ROR - γt were significantly higher than those of the control group, and the differences were statistically significant ( P 〈 0.01 ). Conclusion The imbalance of Th17/Th22/Treg cells may cause threatened abortion.
