摘要
为得到短乳杆菌ZLB004最佳发酵培养基,本试验研究了不同碳源、氮源、缓冲盐、微量元素对短乳杆菌活菌数的影响,并利用响应面分析法对筛选出的主培养基成分进行了优化,得到短乳杆菌ZLB004的发酵培养基为:葡萄糖31.4 g/L、酵母粉3.5 g/L、牛肉膏8.5 g/L、牛肉蛋白胨10 g/L、乙酸钠7.5 g/L、柠檬酸氢二铵3 g/L、磷酸氢二钾3 g/L、硫酸锰0.2 g/L、硫酸锌0.6 g/L、吐温-80 1 ml/L、pH 6.5。短乳杆菌ZLB004在此培养基中37℃培养24 h,活菌数可达3.15×10~9cfu/mL,是优化前活菌数的4.03倍。
To optimize the fermentation medium of Lactobacillus brevis ZLB004,the effects of different carbon source,nitrogen source,buffer salts and microelement on the growth of Lactobacillus brevis were studied.The enrichment medium for Lactobacillus brevis was optimized by response surface methodology,and its composition was:glucose 31.4 g/L,yeast extract 3.5 g/L,beef extract 8.5 g/L,beef peptone 10 g/L,sodium acetate 7.5 g/L,diammonium hydrogen citrate 3 g/L,K_2HPO_4 3 g/L,MnSO_4·5H_2O 0.2 g/L,ZnSO_4·7H_2O 0.6 g/L,Tween 80 1 ml/L,pH 6.5.After cultivated in enrichment medium for 24 h at 37 ℃,the living cells of Lactobacillus brevis ZLB004 were 3.15×10~9cfu/mL,which was about 4.03 times higher than previous optimization.
