摘要
以课题组从龙须菜中分离得到的产琼胶酶弧菌Ag-1为试验菌株,对其产酶培养基和发酵条件进行研究,得到优化后的培养基配方和培养条件为:琼脂0.2%,蛋白胨0.3%,酵母膏0.8%,NaCl 4.0%,MgSO_40.6%,CaCl_20.2%;培养基初始pH 6.5,接种量1%(体积分数),摇床转速240 r/min,装液量50 m L/250 m L,发酵温度33℃,发酵周期24 h。在此条件下做发酵产酶试验,琼胶酶活力达45.51 U/m L,较优化前的7.0 U/m L提高了5.5倍。
A marine agarase-producing strain (Vibrio sp.)Ag-1 was isolated from asparagus and its medium compositions and fermentation conditions for agarase production were studied. The optimal agarase-producing conditions were obtained: agar 0.2%, peptone 0.3%, yeast extract 0.8%, NaCl 4.0%, MgS04 0.6%, CaCl2 0.2%, initial pH of medium 6,5, inoculation volume 1% (v/v), shaking speed 240r/min, medium volume 50mL/250mL, temperature 33 ℃, fermentation time 24 h. Fermenting under these conditions, agarase activity reached 45.51 U/mL, about 5.5 times higher than before optimization (7.0 U/mL).
