摘要
目的:研究白芥子涂法穴位贴敷对哮喘豚鼠的保护作用机制。方法:将豚鼠分为正常组、模型组、贴敷治疗组,OVA致敏,首次激发后,行每周一次的贴敷治疗,共3次。HE染色检测炎症水平、明胶酶谱法检测肺组织金属蛋白酶活性,Western Blot法检测TGFβ、MMP9及Pan-cadherin及磷酸化和非磷酸化Erk1/2、p38的含量。结果:白芥子涂法穴位贴敷能明显减轻哮喘豚鼠的炎症,降低肺组织TGFβ水平并通过抑制Erk1/2及p38MAPK磷酸化,降低肺组织中金属蛋白酶活性、MMP9含量,减少气道上皮屏障关键蛋白Pan-Cadherin破坏。结论:白芥子涂法穴位贴敷可能是通过降低TGFβ-Erk1/2、p38途径抑制金属蛋白酶表达和活性,保护胞外基质细胞连接蛋白,起到防治过敏性支气管哮喘的作用。
Objective: To study the protective mechanism of acupoint application of mustard seed applying on asthmatic guinea pigs. Methods: Guinea pigs were divided into normal group,asthmatic group,paste treatment group,OVA sensitization,and once a week for the first time. The guinea pigs were treated three times. The expression of TGFβ,MMP9,Pan-cadherin and Phosphorylation of Erk1/2 and p38 in lung tissue were detected by Western blot. Results: The treatment of acupoint applying can reduce the inflammation and airway remodeling in asthmatic guinea pigs,and decrease the activities of metalloproteinases,the contents of TGFβ and MMP9 and inhibit the destruction of Pan-Cadherin. Conclusion: Acupoint applying of mustard seed can protect the extracellular matrix of connective tissue through inhibiting the expression and activity of metalloproteinases of TGFβ. It can prevent the airway remodeling of allergic bronchial asthma.
