摘要
目的:从动物实验角度明确医院自拟抗癌扶正协定方对肺癌的药效作用,裨益临床的治疗。方法:本实验考察了抗癌扶正协定方和与肺癌特效化疗顺铂联合作用对小鼠一般体征和生命周期的观察、对移植瘤C57BL/6小鼠体质量、肿瘤体积和重量/肺脏指数、白细胞、红细胞和肺组织的影响。结果:(1)单纯用中药组的小鼠体征未见明显异常,而醇提联合顺铂组小鼠一般体征最差,醇提物联合顺铂组对延长小鼠生命期的效果最差。(2)由于肿瘤的不断增大,模型组、醇提组小鼠体质量也呈增长趋势,但抗癌扶正协定方醇提物逐渐发挥抗癌疗效,肿瘤生长减慢,体质量增长减慢;顺铂有不良反应,小鼠体质量短期内下降较明显。醇提物与顺铂联用并未体现增效减毒功效,反而产生拮抗作用。(3)顺铂和醇提物均具有抑瘤作用,但以顺铂疗效较佳。醇提物联合顺铂治疗后醇提物联合顺铂产生明显拮抗作用。(4)由于肺脏指数=肺重量/小鼠体质量,醇提联合顺铂组小鼠肺脏指数最小,说明肺萎缩程度最严重。可见,顺铂抗癌效果最佳,但当与醇提物联用时,产生拮抗作用,加重不良反应,失去抗肿瘤疗效。(5)各组小鼠RBC数目比较,没有明显差距;对于WBC数目,顺铂组、醇提组WBC数量多,与模型组比较,有显著性差异(P<0.05);醇提物联合顺铂组小鼠WBC数量少,与模型组比较,有显著性差异(P<0.01);醇提物联合顺铂使用,白细胞数目显著降低,可见醇提物会显著增强顺铂毒性,产生较大的白细胞杀伤作用。(6)各图片直观比较结果:模型组癌细胞最多,且出现成块聚集现象,染色深,观察明显。3个治疗组相比,顺铂组癌细胞数目最少,醇提组居中,醇提取物联合顺铂组较多。癌细胞数目直观的反应顺铂具有较强的抗癌活性,顺铂与醇提物联合使用时,抗癌活性下降。结论:抗癌扶正协定方醇提取物有明显的肿瘤抑制作用,但对白细胞也有一定的影响。
Objective: To determine the effect of Fuzheng Anticancer Prescription of our hospital with lung cancer from the perspective of animal experiments,clinical treatment benefits. Methods: This experiment investigated the Anticancer Prescription and lung cancer and the effects of Fuzheng chemotherapy combined with cisplatin,to observe the effect on general signs and life cycle effects on mice transplanted C57BL/6 mice weight and tumor the volume and weight/lung index,white blood cells,red blood cells and lung tissue. Results:(1) with Chinese traditional medicine group of mice showed no abnormal signs,and alcohol extraction combined with cisplatin group were general signs of the worst,alcohol extract and cisplatin group was the worst to prolong the life of the effect in mice.(2) Due to the growing tumor model group,alcohol group,body weight of mice also showed a rising trend,but the prescription alcohol extract anti-cancer Fuzheng gradually play antitumor effect,tumor growth slowed down,slow increase of body weight; toxic side effects of cisplatin,the body weight of mice decreased significantly in the short term. The combination of ethanol extract and cisplatin did not reflect the efficiency and toxicity,but had antagonistic effect.(3)Cisplatin and ethanol extracts have antitumor effect,but the curative effect is better. Ethanol extraction combined with cisplatin in the treatment of alcohol extraction combined with cisplatin has a significant antagonistic effect.(4)Due to lung index = lung weight/body weight of mice,the lung index of mice with ethanol extraction combined with cisplatin was the smallest,which showed that the degree of lung atrophy was the most serious. It is showed that the anticancer effect of cisplatin is the best,but when combined with the alcohol extraction,it can produce antagonism,aggravate the side effects,and lose the anti-tumor effect. Comparison of 5 mice in each group the number of RBC,no significant difference; for the number of WBC,cisplatin group,alcohol group WBC number,compared with the model group,there was significant difference( P〈0. 05); alcohol amount combined with cisplatin in WBC mice,compared with the model group,there was significant difference( P〈0. 01); alcohol extract combined with cisplatin,significantly reduced the number of white blood cells,visible alcohol extract can significantly enhance cisplatin toxicity,cytotoxicity of large white blood cells.(6)The results of visual comparison: model group of cancer cells at most,and the emergence of a block aggregation phenomenon,dyeing depth,observation. Compared to3 treatment groups,the number of cancer cells in the cisplatin group was the least,the alcohol extraction group was in the middle,and the alcohol extract combined with the cisplatin group was more. The number of cancer cells directly response to cisplatin has a strong anti-cancer activity,the use of cisplatin and ethanol extract,the anti-cancer activity decreased. Conclusion: the ethanol extract from the anti cancer and anti-cancer decoction has a significant effect on the cancer,but it also has a certain effect on the white blood cells.
