摘要
目的:观察针灸与振动训练对于缺血缺氧性脑病鼠炎症反应的影响,初步探究针灸和振动训练治疗缺血缺氧性脑病的作用机制。方法:将成功建立缺血缺氧性脑病模型的60只SD大鼠,根据数字随机表法分为4组,即模型对照组(不接受治疗)、针刺组(仅接受针刺治疗)、训练组(仅接受振动训练)以及联合组(针刺联合振动训练)各15只,在建模前、建模后24h以及干预后2周时分别对大鼠神经功能进行评分,同时应用ELISA法检测血清IL-1、IL-6表达水平,应用RT-PCR检测脑组织中TNF-αmRNA、NF-κB mRNA表达水平,同时利用SPSS 19.0对实验结果进行统计分析。结果:建模后,各组大鼠的神经行为评分、血清IL-1、IL-6、脑组织TNF-αmRNA、NF-κB mRNA显著高于建模前(P<0.05),但组间数据无明显差异(P>0.05);干预后,联合组、针刺组、训练组神经行为评分、血清IL-1、IL-6、脑组织TNF-αmRNA、NF-κB mRNA均明显下降,且联合组各指标下降最显著,其次为针刺组,在干预2周时,各组上述指标差异显著(P<0.05)。结论:针灸联合振动训练能够明显减轻缺血缺氧性脑病细胞炎症反应,其作用机制考虑与抑制IL-1、IL-6、TNF-α以及NF-κB等炎症因子表达有关。
Objective: To observe the effect of acupuncture and vibration training on theinflammatory response in hypoxic ischemic encephalopathy ratsandexplore the mechanism of acupuncture and vibration training in the treatment of ischemic hypoxic encephalopathy. Methods: Sixty SD rats of hypoxia ischemia encephalopathy model were established successfully. According to figures,they were randomly divided into 4 groups: model control group( no treatment),acupuncture group( received acupuncture treatment) and training group( received vibration training) and combined group( acupuncture combined with the vibration training). Each group had 15 rats. The neurological function of the rats was evaluated before and after the model 24 h and 2 weeks after the intervention. At the same time,the expression levels of IL-1 and IL-6 in serum was detected by ELISA method. Detect mRNA expression in brain tissue with TNF-α and NF-κB mRNA by using RT-PCRand the statistical analysis of experimental results with SPSS 19. 0. Results: After modeling,the rats neurobehavioral score,serum IL-1,IL-6,TNF-α mRNA and NF-κB mRNAin brain were significantly higher than that before modeling( P〈0. 05),but there was no significant differences between groups of data( P〉0. 05). The intervention group,acupuncture group and training group's neurobehavioral score and serum IL-1,IL-6,TNF-α mRNA and NF-κB mRNAin brain decreased significantlyand the combined group's each index decreased significantly,followed by acupuncture intervention. In 2 weeks,each of these indicators had significant difference( P〈0. 05). Conclusion: Acupuncture combined with vibration training can significantly reduce the inflammatory response in ischemic hypoxic encephalopathyand its mechanism is related to inhibiting the expressions of IL-1,IL-6,TNF-α mRNA and NF-κB mRNA and other inflammatory factors.
