小肠Crohn's病患者肠黏膜中DLG1和DLG5表达的临床观察

Clinical observation of DLG1 and DLG5 expression in intestinal mucosa of patients with small intestine Crohn's disease

目的探讨小肠型克罗恩病(CD)肠黏膜中DLG1和DLG5的表达以及与临床关联分析。方法收集CD患者小肠手术切除标本45例,另收集15例小肠肿瘤以及右半结肠肿瘤手术标本中正常小肠组织作为对照组,采用免疫组织化学方法研究DLG1和DLG5在CD组患者和对照组小肠黏膜上皮组织中的表达,并进行临床观察分析。结果与对照组比较,CD患者小肠黏膜上皮细胞DLG1主要表达在细胞质中,两组患者DLG1表达部位有明显差别(P<0.01),表达量无明显差别。与对照组比较,CD组患者小肠黏膜上皮细胞DLG5均表达在细胞质中,CD组患者DLG5的表达量明显降低(P<0.01)。DLG1表达部位和DLG5表达量与CD患者临床特征无明显关联。结论 DLG5表达降低可能参与CD的发病过程。

Objective To investigate the expression and clinical relevance analysis of drosophila discs large homo- log 1 （DLG1） and drosophila discs large homolog 5 （DLG5） in intestinal epithelial cells in small intestine Crohn＇s disease（CD） patients. Methods A total of 45 small intestine CD patients which underwent surgical treatments were enrolled. Another 15 patients with small intestine tumor or right colon cancer which underwent radical opera- tion in corresponding period were selected as control. All the small intestine specimen were collected and detected by immuno-histochemistry envision staining. The expression of DLG1 and DLG5 in intestinal epithelial cells of the two groups were observed and clinical analysis was carried out. Results Compared with the control group, the ex- pression of DLG1 was mainly expressed in the cytoplasm of the epithelial cells in small intestine CD patients. The expression location of DLG1 in the two groups were significantly different（ P 〈 0.01 ） , while there was no significant difference in the expression quantity. Compared with the control group, all DLG5 were expressed in the cytoplasm of the epithelial cells in small intestine CD patients, and the expression quantity of DLG5 was significantly lower in CD group（P 〈 0.01 ）. There was no significant correlation between the expression location of DLG1 and the expres- sion quantity of DLG5 and the clinical features of CD patients. Conclusion The decreased expression of DLG5 may be involved in the pathogenesis of CD.
