金属基质蛋白酶-2基因启动子区-735C/T多态性与病毒性脑膜炎易感性及血脑屏障通透性关系的研究 被引量:1

The relationship between polymorphism of-735C/T in MMP-2 gene promoter region and susceptibility to viral meningitis and permeability of blood-brain barrier

摘要 目的探讨金属基质蛋白酶-2(matrix metalloproteinase-2,MMP-2)基因启动子区-735C/T位点的基因多态性与病毒性脑膜炎易感性及血脑屏障(blood-brainbarrier,BBB)通透性关系。方法本研究应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法,测定了186例病毒性脑膜炎患者和202例健康者的MMP-2基因启动子区-735C/T位点的基因多态性,对病毒性脑膜炎组和对照组的基因频率以及不同基因型患者脑脊液白蛋白指数(CALB指数)进行对比分析。结果-735C/T位点基因型频率(CC、CT、TT)在病例组中分别为63.44%、31.72%、4.84%,在对照组分别为49.01%、42.57%、8.42%;病例组的C等位基因频率为79.30%、T等位基因频率为20.70%,对照组则为70.30%、29.70%,两者之间比较差异有统计学意义。而CALB指数在CC组(19.019±6.932),CT+TT组(15.835±6.047),两者比较差异有统计学意义(P=0.002)。结论 MMP-2基因启动子区-735C/T位点多态性可能与病毒性脑膜炎的易感性有关,其中C基因可能是易感基因,而T基因可能是保护基因。且该位点多态性可能通过影响BBB的通透性来影响病毒性脑膜炎的易感性。 Objective To explore the relationship between the matrix metalloproteinase-2(MMP-2)gene promoter region-735C/T genetic polymorphism and susceptibility to viral meningitis and blood brain barrier(BBB)permeability. Methods 186 cases of viral meningitis and 202 healthy subjects were enrolled in the study.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)was adopted to determine-735C/T locus genetic polymorphism in MMP-2gene promoter region,gene frequency in viral meningitis group and control group and CSF albumin index(CALB index)in patients were compared and analyzed. ResultsThe frequencies of CC,CT,TT genotype of-735C/T in patients with viral meningitis were 63.44%,31.72%and 4.84%,respectively,they were 49.01%,42.57%and 8.42%in control group,the difference was statistically significant.The frequencies of C allele and T allele were 79.30%,20.70% in patients,and were70.30%,29.70%in controls,the difference was also statistically significant.The CALB index was(19.019±6.932)in the CC group and(15.835±6.047)in the CT and TT group,the difference was statistically significant(P=0.002). Conclusions-735C/T locus genetic polymorphism in MMP-2gene promoter region may be associated with the susceptibility of viral meningitis.C allele may be contributed to the susceptibility,while T allele may be the protective genotype.This polymorphism may influence the susceptibility by affecting the permeability of BBB.
