摘要 肺动脉高压是发病机制复杂的一种致命性疾病,其典型病理变化是肺动脉血管重构,进而降低血管壁的敏感性及增加肌束的收缩力,引起肺动脉压力升高、肺血管阻力增加,最终导致右心衰竭。多项研究表明,转化生长因子β1(transforming growth factor-β1,TGF-β1)在多种疾病中发挥重要作用,尤其是在心脑血管疾病。TGF-β1参与细胞的增殖、分化、迁移及凋亡等活动,通过调控多种信号通路促进平滑肌细胞增殖、细胞外基质沉积及内皮间质转化(endothelialto-mesenchymal transition,End MT)等过程,从而诱导肺动脉高压的发生发展。该文针对TGF-β1在肺动脉高压中的作用进行综述,从而为肺动脉高压的治疗提供潜在的药物靶点和治疗策略。 Pulmonary artery hypertension is a disease with com-plicated pathogenesis, which is characterized by enhanced pul-monary artery constriction and arterial wall remodeling, leading to progressive increase of pulmonary vascular resistance and pul-monary artery pressure, then resulting in right heart failure. Many studies have shown that transforming growth factor-β1 (TGF-β1 ) plays an important role in the development of various diseases, especially in cardiovascular and cerebrovascular disea-ses. TGF-β1 is involved in multiple cellular responses including cell proliferation, differentiation, migration and apoptosis. TGF- pl participates in pulmonary artery hypertension mainly via pro-moting the proliferation of pulmonary artery smooth muscle cellsas well as inducing the deposition of extracellular matrix and en- dothelial-to-mesenchymal transition( EndMT) through many sig-naling, which is mainly dominated by pulmonary artery smooth muscle cells and pulmonary artery endothelial cells. This review mainly introduces the role of TG F -β1 in pulmonary artery hyper-tension in order to provide potential drug targets and therapeutic strategies for pulmonary artery hypertension.Key words : pulmonary artery hypertension; TGF -β1 ; vascular remodeling; pulmonary arterial smooth muscle cells; extracellu-lar matrix; pulmonary arterial endothelial cells; endothelial-to- mesenchymal transitionas well as inducing the deposition of extracellular matrix and en- dothelial-to-mesenchymal transition( EndMT) through many sig-naling, which is mainly dominated by pulmonary artery smooth muscle cells and pulmonary artery endothelial cells.
