冰冻切片和石蜡切片对免疫组化染色效果的对比分析 被引量:3

Comparison over the Effects of Paraffin Sections and Frozen Sections on Immumohistochemical Staining in the Tissues of Skin and Granulation

摘要 目的研究皮肤、肉芽组织采用石蜡切片以及冰冻切片免疫组化的染色效果。方法 2014年8月—2016年8月期间选取Wistar大鼠共60只,双盲法随机分为A、B两组各30只。实验前对60只大鼠进行背部切割伤,脱毛后在每只大鼠背部切割伤部位取1 cm×1 cm大小的皮肤块作为标本,A组大鼠皮肤块标本采用石蜡制片,B组大鼠皮肤块标本采用冰冻制片。对两组大鼠标本切片进行免疫组化染色,观察两组大鼠标本切片染色情况。结果 B组SP、EGF、EGFR以及FGF强阳性率分别为96.67%、100.00%、96.67%、93.33%,明显高于A组的0.00%(P<0.05)。A1组抗原活性阳性率分别为93.33%、96.67%、93.33%、100.00%。明显高于A2组的13.33%、13.33%、3.33%、6.67%以及A3组的10.00%、16.67%、13.33%、3.33%(P<0.05)。B1组EGF、FGF抗原活性强阳性率分别为100.00%、93.33%,高于B2组的6.67%、3.33%(P<0.05)。结论冰冻制片免疫组化染色效果更佳,值得临床应用及推广。 Objective To s tudy th e effects of paraffin sections and frozen sect io ns on Immumohistochemical s taining in the t issues of skin and granulat ion. Methods 6 0 W is ta r rats were se le c ted from August 2 01 4 to August 2 01 6 and randomly d i -vided into group A and group B with 30 cases in each group respectively. Before experiment, 60 rats were concis on the back. After removing the hair , the skin with the size of 1 cmx1 cm was taken from cut injuries on the back and served as sample. The specimens of rats in group A were adopted paraff in sect ions , and rats in B group were adopted frozen sections. Immunohis tochemical staining was performed on the two groups of rats to observe the sect ion staining of the rats in two groups. Results T he positive ra te s of SP, EGF, and EGFR in group B 9 6 .6 7%、 1 0 0 .0 0%、 9 6 .6 7%、 9 3 .3 3% were significantly higher than those of group A 0.00%(P〈0.05). In group A, the weak positive rates were relatively higher with regard to ant i -gen activi ty detect ion for specimens of A2 and A3 (93.33%、 9 6 .6 7%、 9 3 .3 3%、 100.00% vs 1 3 .3 3%、 1 3 .3 3%、 3 .3 3%、 6.67% vs 15.00%、 16.67%、 1 3 .3 3%、 3 .3 3%) . However, th e above men tion ed an tig en activi ty d e te c t io n ra tes of A1 were significantly higher than those of A2 and A3 with (P〈0.05). In group B, the strong positive rates of B1 group with signif icantly higher than those of B2 with regard to ant igen activi ty detect ion of EGF and FGF with (10.00%、 93.33% vs 6 .6 7%、 3 .3 3% ) (P 〈 0 .0 5 ) . Conclusion T h e effect of frozen sections is s ignif icantly b e t te r th an th a t of para ff in sect ions on immumohis tochemical s ta in -ing, which is worthy of cl inical appl icat ion and popularization.
