6周有氧运动对高脂饲养大鼠性激素及组织芳香化酶表达量的影响 被引量:1

The effect of six weeks aerobic exercise on sex hormone and P450 arom expression of tissues in male rats with high fat diet

摘要 目的:观察高脂膳食对雄性大鼠性激素、性腺和性腺外组织中芳香化酶表达量的影响及自由转轮运动的干预效果,探讨有氧运动影响高脂膳食大鼠性激素水平的可能机制。方法:40只3周龄雄性SD大鼠随机分为4组:普通膳食对照组(C组)、普通膳食运动组(E组)、高脂膳食组(D组)、高脂膳食运动组(Er D组)。E、ErD两组进行自由转轮运动,每天2h,每周训练5天。6周后腹主动脉取血,制备血清,并取睾丸、肝脏、股四头肌和肾脏,检测总睾酮(T)和雌二醇(E_2)含量,并检测组织中芳香化酶(P450 arom)的表达情况。结果:(1)与C组相比,D组大鼠血清T显著降低(P<0.05),E_2显著升高(P<0.05);与D组相比,ErD组大鼠T水平升高,但是无显著性,E_2水平明显下降(P<0.05)。(2)与C组相比,D组大鼠睾丸和股四头肌中P450 arom表达量显著升高(P<0.05),肝脏和肾脏中非常显著性升高(P<0.01);与D组相比,ErD组大鼠睾丸和肾脏中P450 arom表达量显著降低(P<0.01,P<0.05),肝脏和股四头肌中也出现降低,但差异不明显(P>0.05)。结论:高脂膳食可引起雄性大鼠睾酮水平降低和雌二醇水平增高。6周自由转轮运动一定程度上改善了高脂膳食诱发的性激素水平异常现象,其作用可能与降低性腺(睾丸)及性腺外组织芳香化酶的表达有关。 Objective: To investigate the effect and mechanism of high fat diet on sex hormones and expression of P450 arom in gonad and extragonadal tissues and the effect of voluntary wheel running treatment of them. Method: Forty male 3-week-old SD rats were randomly divided into following groups: control group fed with normal diet (C), exercise group fed with normal diet (E),diet group fed with high fat diet (D), exercise and di- et group treated with exercise and high fat diet (ErD). E and ErD groups were assigned to play running wheel freely. 2 times/d, lh/time, 5d/w, 6 weeks in total. 6 weeks later, after the rats had been adequately anesthetized with pentobarbital sodium, blood sample was obtained. Then, testis, liver, quadriceps femoris, and kidney were rapidly excised. Serum T and E2 were determined. And the protein expression of P450 arom was later analyzed. Result: (1)Compared with C group, serum T significantly decreased in D group (P 〈 0.05), and serum E2 obviously increased in D group (P 〈 0.05). Compared with D group, serum T in ErD group was high, but the difference was not significant (P〉0.05), serum E2 significantly decreased in ErD group(P〈 0.05). (~Compared with C group, the expression of P450 arom in testis and quadriceps femoris were significantly higher (P〈 0.05) in D group, the expression of P450 arom in liver and kidney were very significantly higher (P〈 0.01); Com- pared with D group, the expression of P450 arom in testis and kidney significantly decreased (P〈 0.01, P〈 0.05) in the ErD group, the expression of P450 arom in liver and quadriceps femoris also decreased, but the difference was not significant (P 〉 0.05). Conclusion: High fat diet induces the unbalance of T and E2. 6 weeks voluntary wheel running improves the phenomenon of the metabolism disorder of T and E2 induced by high fat diet at certain extent, which may be involved in the decrease of the P450 arom protein in gonad and extragonadal tissues.
