摘要
代谢组学是利用高通量检测技术对内源性代谢物进行定性和定量分析,从而挖掘代谢物与相应生物学变化的相对关系。目前,代谢组学在药物研发、疾病研究、药物毒理学等领域发挥着越来越大的作用。液相-质谱联用平台,检测的相对分子质量范围大、分辨能力强、扫描速度快,拥有很高的灵敏度和较宽的动态范围,其在代谢组学中的应用得到越来越多研究者的认可。生物样品的采集和制备是代谢组学试验中至关重要的步骤,直接决定着试验结果的可靠性。由于样本自身的复杂性以及对样本采集和制备过程中细节的忽视,往往导致试验结果出现偏差,甚至导致错误的结论。因此充分验证样本的采集和处理方法在试验中尤为重要。本文主要综述了基于质谱平台的代谢组学常用生物样本的采集和制备方法,为代谢组学试验提供相关参考。
Metabonomics could make qualitative and quantitative analysis of endogenous metabolites with the application of high-through test technology, and thus dig out relative relationship between metabolites and the relevant biological change. Currently,metabonomics play an important role in many research fields including drug discovery, disease research, drug toxicology and et al. Liquid chromatography-mass spectrometry (LC-MS) platform has a wide detection range of molecule weight, high resolution ability, fast scan speed, high sensitivity and wide dynamic range. The appli- cation of LC-MS platform in metabonomic study has benn gradually accepted by many research- ers. The collection and preparation of biological sample are crucial steps in metabonomic experi- ment, which directly determine the reliability of the experimental results. The complexity of sam- ple itself and the ignoring details in sample collection and preparation process frequently cause de- viations in experimental results, and even erroneous conclusion. Therefore,it is essential to validate the method of sample collection and preparation. This article mainly reviewed the method of sam- ple collection and preparation for metabonomic experiment with LC-MS platform, which could pro- vide reference for related research.
