摘要
目的观察酒精性肝损伤模型大鼠肝功能动态变化规律及乌药对酒精性肝损伤的干预作用。方法将33只SD大鼠随机分为空白对照组、模型组和乌药干预组,各11只,各组大鼠分别于实验第14、21、33天眼眶静脉丛取血,测定血清丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)水平。结果模型组大鼠14、21、33天的ALT水平分别为(70.8±8.5)U/L、(74.8±10.0)U/L、(86.5±19.3)U/L;AST水平变化不显著,分别为(185.9±24.5)U/L、(175.1±15.5)U/L和(177.9±42.7)U/L。模型组21、33天的ALT、AST与空白对照组[ALT:(58.4±13.0)U/L、(50.1±6.1)U/L;AST:(175.1±15.5)U/L、(137.7±20.7)U/L]比较,差异有统计学意义(P<0.01,P<0.05);与模型组比较,乌药干预组第21、33天ALT分别为(62.6±9.6)U/L和(65.8±12.6)U/L,AST分别为(142.6±25.0)U/L和(138.7±18.2)U/L,差异有统计学意义(P<0.05,P<0.01)。结论酒精性肝损伤模型大鼠肝损伤严重程度随着造模时间延长逐渐加重;乌药对酒精引起的模型大鼠肝损伤有保护作用。
Objective To observe the changes of liver function of rats with alcoholic liver disease and the intervention effect of combined spicebush root on the changes. Methods Thirty-three SD rats were randomly divided into three groups (n=11): control group, model group, and treatment group. Except for control group, the rat model of alcoholic liver disease was established. On experimental day 14, 21, and 33 the blood samples were drawn from rats" orbital vein to detect alanine aminotransferase (ALT) and aspartate transaminase (AST) for evaluation of liver function. Results In model group, ALT on day 14, 21, and 33 were 70.8±8.5U/L, 74.8±10.0U/L, 86.5±19.3U/L, respectively, and AST on day 14, 21, and 33 remained fast stable: 185.9±24.5U/L, 175.1±15.5U/L, and 177.9± 42.7U/L. The ALT and AST on day 21 and 33 in model group were significantly higher than those in control group(ALT: 58.4+13.0U/L and 50.1±6.1U/L; AST: 175.1±15.5U/L and 137.7±20.7U/L; P〈0.01 or P〈0.05). The ALT and AST on day 21 and 33 in treatment group (ALT: 62.6±9.6U/L and 65.8±12.6U/L; AST: 142.6±25.0U/L and 138.7±18.2U/L) were significantly decreased compared with model group (P〈0.01 or P〈0.05). Conclusion The severity of live dysfunction increases with the days of molding and combined spicebush root can reverse the dysfunction in certain degree.
