摘要
针对沙棘叶的现有提取工艺采用单因素试验法进行优化,并分别利用紫外分光光度法和HPLC法测定总黄酮含量和分类黄酮含量,建立了HPLC同时测定五种黄酮品类的定性定量方法,并依据检测数据最终确定了沙棘最优提取工艺条件:70%的乙醇在80℃提取,料液比(g/mL)为1∶16,单次提取时间为2 h,提取3次,获得了较高的黄酮提取率,并实现了分类黄酮提取的条件优化。
The method of single factor experiment was optimized by single factor test. The content of total flavonoids and the content of different flavonoids were determined by ultraviolet spectrophotometry and HPLC, respectively. A qualitative and quantitative method for simultaneous determination of five flavonoids was established by HPLC. The optimal extraction conditions of seabuckthorn were determined according to the test data: 70% ethanol was used as extraction solvent, extraction temperature was 80℃, the ratio of material to liquid (g/mL) was 1 : 16, the extraction time was 2 h, extracted three times. High flavonoid extraction rates were achieved and the different flavonoids were optimized for the extraction conditions.
