摘要
利用纤维素酶辅助黑曲霉提取柠檬皮中的果胶,采用咔唑硫酸法测定提取液中的果胶含量。在单因素实验的基础上,以果胶提取率为指标,利用Box-Behnken设计四因素三水平进行响应面试验,建立各因素与响应值之间的数学模型,确定最佳提取工艺。结果表明,最佳工艺条件为:纤维素酶处理时间1.0 h,处理温度45℃,缓冲液pH=5.0,酶量0.5%,发酵时间24 h,在此工艺条件下,果胶提取率为7.38%。
Pection was extracted by aspergillus niger assisted with cellulase from lemon peel. The content of pectin was measured by carbazole-sulfuric acid method. A Box-Behnken design involving four factors at three levels each was used to determine the optimum extraction conditions using pectin yield as response. The optimum extraction conditions were obtained as follows: cellulase treatment time was 1.0 h, cellulase treatment temperature was 45℃, buffer pH = 5.0, cellulase concentration was 0. 5% and fermentation time was 24 h. Under the optimized conditions, the yield of pectin was 7. 38%.
