双齿围沙蚕消化道降油细菌的分离鉴定及降油性能研究

摘要 从采自辽宁大连地区的双齿围沙蚕消化道分离得到3株能利用柴油为唯一碳源的石油降解菌,编号分别为,编号为SC11-32-2,SC11-37,SC11-38。通过形态学和16S rDNA序列比对对菌株进行鉴定,结果显示:SC11-32-2菌株16S rDNA序列与Pseudomonas monteilii同源性为99.9%,SC11-37菌株16S rDNA序列与Vibrio alginolyticus同源性为99%,SC11-38菌株16S rDNA序列与Pseudomonas composti同源性99%。继而测定了不同浓度(10~7cfu/mL、10~8cfu/mL、10~9cfu/mL)分离菌对柴油的降解率,以及加入葡萄糖后不同浓度(10~7cfu/mL、10~8cfu/mL、10~9cfu/mL)分离菌对柴油的降解率。结果显示:SC11-32-2菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为26.31%、44.52%、51.26%;SC11-37菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为7.89%、19.22%、23.36%;SC11-38菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为12.32%、27.33%、35.56%;加入终浓度为4g/L葡萄糖后,3株菌对柴油的降解率均有显著提高,SC11-32-2菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为87.56%、88.33%、88.62%;SC11-37菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为67.88%、69.12%、70.38%;菌株在10~7cfu/mL、10~8cfu/mL、10~9cfu/mL接种量条件下的降油率分别为75.67%、78.35%、79.56%。 In this study, 3 strains of oil-utilizing bacteria were isolated from the digestive tract of Pernereis aibuhitensis collected from DaLian, Liaoning, China through medium containing diesel oil as a sole source of carbon., named SC11-32-2, SC11-37, SC11-38. Cellular morphological observations and molecular indentifications of 16S rDNA were used to identify strains SCll-32-2, SCll-37 and SCll-38. The results showed that strain SC11-32-2 belonged to the genus Pseudomonas monteilii, and the homology is 100%;strain SC11-37 belonged to the genus Vibrio alginolyticus, and the homology is 99%;strain SC11-32-2 belonged to the genus Pseudomonas composti, and the homology is 99%.The initial diesel oil-degrading efficiency of different inoculation quantity was detected, meanwhile the oil- degrading efficiency was studied when glucose was added. The results showed that the initial diesel oil-degrading efficiency of SC11-32-2 in the inoculation quantity of 10-7cfu/mL, 10-8cfu/mL, 10-9cfu/mL was 26.31%, 44.52% and 51.26%, respectively. And significantly increased to 87.56%, 88.33% and 88.62% after adding 4 g/L glucose, In the same situation, the diesel oil-degrading efficiency of SC11-37 increased from 7.89%,19.22% and 23.36% to 67.88%, 69.12% and 70.38% in three inoculation levels. The diesel oil-degrading efficiency of SCll-38 increased from 12.32%, 27.33% and 35.56% to 75.67%, 78.35% and 79.56% in three inoculation levels.
