摘要
目的:探讨系统鉴定法对新疆和田市售维药材赤芍的精准鉴别,为保证药材来源的准确性提供科学依据。方法:应用系统鉴定法,即综合性状、显微鉴别(横切面、粉末)和DNA条形码分析等方法,配合Gen Bank数据库、NCBI的blast功能和Contig Express、DNAMAN、Editseq、Me GA5等软件的使用,对维药材赤芍的性状、显微及ITS序列特征进行分析,并且对块根芍药及其易混品种进行ITS序列分析、遗传距离、特异位点、N-J系统聚类树分析。结果:通过系统鉴定法有效鉴别出维药材赤芍的药材来源为块根芍药Paeonia hybrida Pall。并且应用ITS序列能快速有效区分块根芍药与窄叶芍药和新疆芍药。结论:本研究证实系统鉴定法能够完成维药材克孜力乔古鲁克的精准鉴别,为确保其准确的药材来源提供科学依据,对块根芍药的深入研究具有重要意义。
Objective: To complete the accurate identification of Paeonia Radix Rubra (PRR) of Uygur medicine herbal purchased from medicine market of Hotan Xingjiang, systematic identification method was adopt- ed to provide a scientific basis for ensuring the accuracy of material sources. Methods: The morphological, mi- croscopic and characteristics of ITS sequence were analyzed by systematic identification method which comprise morphological identification, microscopic identification and DNA identification method combined with database of GenBank, function of blast in National Center for Biotechnology Information (NCBI), a series of software of Con- tigExpress, DNAMAN, Editseq, MeGA5. Despite all that, Interspecific and intraspecific Kimura 2-parameter(K 2- P) genetic distance, specific site and Neighbor-joining(N-J)system cluster tree were also analyzed between Paeo- nia. anomala var intermedia and its adulterants. Results: This study successfully identified that PRR of Uygur medicine herbal rooted in Paeonia. hybrida Pall and distinguished it between hybrida, P. anomala and P. sin- jiangensis. Conclusion: Systematic identification method could be used for accurate identification of PRR of Uygur medicine herbal and lay the foundation for popularization and application of it, have important implication for further research and development of P. hybrida.
