中药复方对难治性癫痫大鼠多药耐药P-糖蛋白表达的影响

Effect of Chinese Herbal Compound on Expression of Multi-drug Resistance Protein P-glycoprotein for Refractory Epilepsy

目的:探讨中药复方熄风胶囊对锂-匹罗卡品致难治性癫痫模型大鼠多药耐药蛋白P-糖蛋白(P-gp)表达的影响。方法:建立氯化锂-匹罗卡品癫痫大鼠模型。实验大鼠随机分为8组:正常对照组(空白组)、模型对照组(模型组)、熄风胶囊低剂量治疗组(熄低组)、熄风胶囊中剂量治疗组(熄中组)、熄风胶囊高剂量治疗组(熄高组)、卡马西平治疗组(CBZ组)、熄风胶囊中剂量+卡马西平治疗组(熄卡组)、熄风胶囊中剂量+1/2卡马西平治疗组(熄卡低组)。熄低、中、高组分别予熄风胶囊0.33g、0.66g、0.99g,浓缩剂2ml;CBZ组予CBZ 20mg/kg;熄卡、熄卡低组分别予熄风胶囊0.66g、0.33g浓缩剂2ml和CBZ 20mg/kg;模型组和空白组分别予生理盐水2ml。每天上午灌胃一次,共持续60天。给药结束后检测各组大鼠P-gp蛋白的表达。结果:与空白组比较,模型组P-gp的表达高于空白组(P<0.05),各治疗组P-gp的表达均低于空白组(P<0.05);与模型组比较,各治疗组P-gp的表达均低于模型组(P<0.05);与CBZ组比较,熄低组、熄卡低组P-gp的表达均低于CBZ组(P<0.05);中药组各组之间均没有显著差异(P>0.05);熄卡低组P-gp的表达低于熄卡组(P<0.05)。结论:熄风胶囊对癫痫大鼠脑组织P-gp的表达有显著抑制作用,与剂量无相关性;与CBZ联合治疗,对IE大鼠脑组织P-gp表达均有显著抑制作用,效果优于单纯CBZ组。

Objective:To study the different expression of multidrug transporter P-gp in the brain tissue of lithium chloride pilocarpine epileptic rats being treated by the Chinese herbal compound Xifeng Capsule.Methods:Established the lithium chloride-pilocarpine epileptic rat model.All rats were randomly divided into eight groups:normal controlled group,model controlled group,Xifeng Capsule low-dose treatment group(Xifeng low-dose group),Xifeng capsule middle-dose treatment group(Xifeng middle-dose group),Xifeng capsule high-dose treatment group(Xifeng high-dose group),carbamazepine treatment group(CBZ group),Xifeng middle-dose+carbamazepine in the treatment group(Xi-car group),Xifeng Capsule middle-dose+1/2dose carbamazepine treatment(Xi-car low group).Xifeng low,medium and high group respectively given 0.33g,0.66g and 0.99g,thickener 2ml;CBZ dose group given CBZ 20 mg/kg;the Xi-car middle-dose and Xi-car low-dose group respectively were given Xifeng Capsule 0.66g and 0.33g thickener 2ml and CBZ 20mg/kg;the normal controlled group and the model controlled group respectively given 2ml saline solution.Morning lavage once a day,a total of 60 days.Then tested the P-gp expression of each group.Results:The Western-blot results showed that the expression of P-gp in the model group was higher than that in the normal group(P<0.05),while the expression of P-gp in all the treatment groups were lower than that in the normal group(P<0.05);the expression of P-gp in all the treatment groups were all lower than that in model group(P<0.05);compared with the CBZ group,the expression of P-gp in Xifeng low-dose group and Xifeng capsule middle-dose+1/2 dose CBZ group were each lower than that in the CBZ group(P<0.05);among the traditional Chinese medicine groups,they had no statistical differences from each other(P>0.05);the expression of P-gp in Xifeng Capsule middle-dose+1/2 CBZ group was lower than that in Xifeng Capsule middle-dose+CBZ group(P>0.05).Conclusion:Western-blot showes that Xifeng Capsule can significantly reduce the expression of P-gp in the brain issue of IE rats,and dose not correlation;associated with CBZ therapy,the expression of P-gp IE rats have significantly inhibitory effect,and it is better than that of pure CBZ group.
