摘要 通过HPLC标准曲线法进行酵母中还原型和氧化型谷胱甘肽含量测定的研究,利用Intersil ODS-3 C_(18)柱,以庚烷磺酸钠-磷酸盐缓冲溶液(pH3.0)和甲醇(v∶v=90∶10)为流动相,流速为1.0 mL/min,检测波长为210 nm,得到氧化型和还原型谷胱甘肽的相关系数均为0.9999,检测限分别为2.40 mg/L、2.27 mg/L,定量限分别为9.0 mg/L、8.5 mg/L,精密度分别为2.00%、0.94%,回收率分别为100.12%~103.43%和95.23%~96.44%之间。 The content of reduced/oxidized glutathione in yeast was measured by HPLC standard curve method. Intersil ODS-3 C 18 column was applied, sodium heptane sulfonate-hydrogen phosphate buffer solution (pH=3) and methanol (v: v=90:10) was used as mobile phase, flowing rate was 1.0 mL/min, and detection wavelength was 210 nm. The correlation coefficient of reduced/oxidized glutathione was 0.9999,and their detection limits were 2.27 mg/L and 2.40 mg/L respectively, their quantification limits were 8.5 mg/L and 9.0 mg/L respectively, their precision were 0.94 % and 2.00 % respectively, and their recoveries were 95.23 % to 96.44 % and 100.12 % to 103.43 % respectively.
