SIRT2在膀胱尿路上皮癌中的表达及意义

Expression and clinical significance of SIRT2 in transitional cell carcinoma of bladder

目的:研究SIRT2在人膀胱尿路上皮癌中的表达及临床意义,探讨其在肿瘤发生发展中的作用。方法:收集人正常膀胱移行上皮组织标本16例,膀胱尿路上皮癌标本36例。按照病理分级、临床分期分组,用免疫组织化学法(SP法)检测SIRT2蛋白的表达,分析其与膀胱尿路上皮癌的病理分级、临床分期的关系。结果:正常膀胱移行上皮组织中SIRT2阳性表达率为87.50%;膀胱尿路上皮癌组织中SIRT2阳性表达率为55.56%,膀胱尿路上皮癌组织中的SIRT2表达水平明显低于正常膀胱移行上皮组织(P=0.04)。SIRT2蛋白在膀胱尿路上皮癌中低级别组与高级别组阳性表达率分别为75.0%和40.0%,差异有统计学意义(P=0.04),同时SIRT2在非肌层浸润(T_1)与肌层浸润(T_(2-4))间阳性表达率分别为78.6%及40.9%,差异有统计学意义(P=0.03)。结论:SIRT2蛋白在膀胱尿路上皮癌组织中的表达明显低于正常膀胱移行上皮组织,且与膀胱尿路上皮癌的病理分级、临床分期呈负相关,提示SIRT2蛋白表达下调在膀胱尿路上皮癌发生、发展中起重要作用。

Objective：To study the expression and clinical significance of SIRT2 in transitional cell carcinoma of bladder （TCCB） , and to explore its role in progression and invasion of TCCB. Methods ： A total of 36 patients with pathological identified TCCB undergoing radical cysteetomy in our hospital during March 2013 to March 2015 were re- cruited in this study. Their clinical data including pathological grades and clinical stages were collected. The expres- sion of SIRT2 in the tissue samples of 16 normal bladder tissues and these 36 TCCB were tested by SP immunohisto- chemistry. The relationships between SIRT2 with pathological grades and clinical stages were also analyzed. Results： The expression rate of SIRT2 in the tissues of transitional epithelium of normal bladder was 87.50% ,and the expres- sion rate of SIRT2 in the tissues of TCCB was 55.56%. The expression of SIRT2 in TCCB was significantly lower than that in normal bladder tissues （P = 0.04）. In the tissues of TCCB, the expression rate of SIRT2 in low - grade and high grade were 75.0% and 40.0% , respectively. The difference was statistically significant （P = 0.04）. The expres- sion rate of SIRT2 in nonmuscle - invasion and muscle - invasive tumors were 78.6% and 40.9% ,respectively. Sig- nificant difference was found between nonmuscle - invasion and muscle - invasive tumors （ P = 0.03 ）. Conclusion： The expression of SIRT2 in TCCB is significantly lower than that in normal bladder tissues and negatively correlatedwith pathological grades and clinical stages,indicating that SIRT2 plays an important role in the occurrence and devel- opment of TCCB.
