摘要
目的:探讨穿山甲炮制增效的机理。方法:采用TOF MS-IDA-MS/MS对穿山甲炮制前后成分变化进行研究,Waters C18色谱柱(2.1mm×150mm,1.7μm);柱温40℃。流动相为0min,乙腈-水(5∶95),30min乙腈-水(95∶5);体积流量0.4 ml/min。质谱定性采用电喷雾离子源(ESI)的飞行时间质谱,负离子模式下采集数据,利用Markerview分析软件对化合物进行筛选,通过精确相对分子质量和二级质谱裂解碎片鉴定监测到的化学成分。结果:经过PCA分析,可以很好地区分穿山甲炮制前后成分的变化,穿山甲经炮制后,新增丝-酪环二肽及苯丙-脯环二肽等四种成分。结论:穿山甲经高温炮制后环二肽等亲水性成分含量显著增加。
Objective To conduct a preliminary research on the synergistic mechanism of pangolin scales.Methods:The components of pangolin scales were analyzed by TOF MS-IDA-MS/MS before and after processing.The target compounds were also analyzed using analysis software such as Markerview. The compounds were thenscreened and the chemical constituents detected by the relative molecular mass and MS2 were identified. Results:The results have shown that PCA analysis conducted was able to effectively distinguish the changes that occurred beforeand after processing pangolin scales. After undergoing processing, four (4) new componon the level of Cyclo-(Ser-Tyr) and Cyclo-(Phe-Pro) , two components already present in pangolin scales. Conclu-sion: The Hydrophilic substances present on the cyclo-dipeptide are increased after processidrophilic contents of cyclo-dipeptide are known to be an important material basis of synergism of pangolin scales.
