参黄洗液对大鼠痔术后创面修复过程中血清IL-8及TNF-α的干预作用

Impacts on IL-8 and TNF-α in the Intervention with Shenhuang Lotion for Wound Repair after Hemorrhoids Operation in the Rats

目的研究参黄洗液对大鼠痔术后创面修复过程中白介素-8(IL-8)及肿瘤坏死因子-α(TNF-α)的干预作用,探讨参黄洗液促痔术后创面修复的部分免疫学机制。方法 90只SD大鼠,随机分成空白对照组(A)、模型对照组(B)、金玄痔科熏洗散组(C)、参黄洗液低剂量组(D)、参黄洗液中剂量组(E)、参黄洗液高剂量组(F)。除空白对照组外,其他组均采用肛周局部注射冰醋酸造模成痔术后感染创面模型,各治疗组分别给予相应的药物熏洗,模型对照组及空白对照组给予生理盐水熏洗,连续治疗1周,治疗第3天、第7天分别进行眼眶采血,用酶联免疫法检测血清中IL-8及TNF-α的含量。结果与A组比较,B组及各治疗组大鼠治疗后第3、7天血清中IL-8、TNF-α含量均有上升(P<0.01,P<0.05),说明痔术后创面感染能加重机体炎症反应;与B组比较,各治疗组大鼠治疗后第3、7天血清中IL-8、TNF-α的含量均有下降,其中F组下降最为明显(P<0.01),说明参黄洗液熏洗坐浴能控制痔术后创面感染,减轻机体炎症反应。治疗后第7天各治疗组大鼠血清中IL-8、TNF-α的含量较本组第3天明显下降。结论参黄洗液熏洗促进痔术后创面愈合机制可能与下调大鼠血清中IL-8及TNF-α水平有关。

Objective To study the impacts on IL -8 and TNF -α in the intervention with shen- huang lotion for wound repair after hemorrhoids operation in the rats and discuss the partial immunological mechanism on the impacts. Methods Ninety SD rats were randomized into a blank control group（ A group）, a model control group（ B group） , a j inxuan zhike xunxi powder group（ C group）, a low- dose shenhuang lotion group（ D group） ,a middle - dose shenhuang lotion group（ E group） and a high - dose shenhuang lotion group（ F group）. Except in the A group,the ice acetic acid was injected locally to prepare the model of infectious wound of hemorrhoids operation in the rats of the rest groups. The correspond medicinal fumigations were applied in the treatment groups. The fumigation with physical saline was used in the A and B groups. The treatment lasted 1 weeks. On the 3^rd and 7^th days of treatment, the orbital blood was collected. The enzyme - linked immunoassay was adopted to determine the levels of IL - 8 and TNF - α in the serum. Results Compared with the A group, the IL -8 and TNF -α were increased on the 3^rd and 7^th days after treatment in the B group and the treatment groups（ P 〈 0.01 ,P 〈 0.05 ） ,indicating that the wound infection after hemorrhoids operation got the body inflammatory reactions worse. Compared with the B group, the IL - 8 and TNF - α were reduced on the 34 and Th days after treatment in all of the treatment groups, of which, the reducing was the most apparent in the F group（ P 〈 0. 01 ） ,explaining that the bath therapy with shenhuang lotion controlled the wound infection after operation and alleviated the body inflammatory reactions. The levels of IL -8 and TNF -α on the 7^th day after treatment in every treatment group were all reduced apparently in the self- comparison with those on the 3^rd day. Conclusion Shenhuang lotion promotes wound healing after hemorrhoids operation,which is possibly relevant with the down- regulation of serum IL-8 and TNF- α in the rats.
