摘要
目的:研究头花蓼中木脂素类成分及其降糖活性。方法:采用硅胶柱层析、反相ODS柱色谱、Sephadex LH-20等分离手段对头花蓼降糖活性部位进行分离纯化,通过波谱数据分析进行结构鉴定。采用体外α-淀粉酶抑制实验对分离得到的木脂素类成分进行体外活性实验。结果:从头花蓼中共分离出7个木脂素类成分,分别鉴定为:(+)异落叶松脂醇(1)、(-)-南烛木树脂酚-2a-O-[6-O-(4-羟基-3,5-二甲氧基)-苯甲酰基]-β-D-葡萄糖苷(2)、(-)-异落叶松脂素-2a-O-β-D-木吡喃糖苷(3)、(+)-5'-甲氧基异落叶松脂素-9-O-β-D-木吡喃糖苷(4)、(-)-异落叶松脂素-3a-O-β-D-葡萄糖苷(5)、nudiposide(6)、lyoniside(7)。结论:其中,化合物1、2、5、7为首次从该植物中分离得到,化合物2、7为首次从蓼属植物中分离得到。化合物1、2、4显示出了较好的体外α-淀粉酶抑制活性,其中化合物2对体外α-淀粉酶抑制作用显著。
Objective :To study lignans of Polygonum capitatum and their hypoglycemic active constituents. Methods:Lignans were separated and purified by silica gel column chromatography,reversed phase ODS chromatography and Sephadex LH-20. The compounds were identified by their spectral data, and α-amylase inhibitory test were used to evaluate the hypoglycemic activities of the compounds, Results:Seven lignans were isolated from Polygonum capitatum. They were identified as ( + ) -isolariciresinol ( 1 ), ( - ) -lyo- niresinol-2a-O-[ 6-O-( 4-hydroxy-3,5-dimethoxy ) -benzoyl ] -β-D-glucopyranoside ( 2 ), ( - ) -isolariciresinol-2a-O-β-D-xylopyranoside (3), ( + )-5'-methoxyisolariciresinol-9-O-β-D-xylopyranoside (4), ( - )-isolariciresinol-3a-O-^-D-glucopyranoside (5), nudiposide (6) ,lyoniside (7). Conclusion:Compounds 1,2,5,7 are isolated from this plant for the first time, and compounds 2 and 7 are separated from Polygonum genus for the first time. Compounds 1,2, and 4 shows good a-amylase inhibitory activity in vitro, and compound 2 exhibits remarkable a-amylase inhibitory effect.
