补阴消郁汤配方颗粒对更年期小鼠抑郁的抑制作用研究

目的:考察补阴消郁汤配方颗粒对更年期模型小鼠抑郁行为的保护作用,并考察其可能的机制。方法:通过卵巢摘除建立更年期模型,补阴消郁汤配方颗粒2g/kg、6g/kg、18g/kg连续给药4周,通过小鼠旷场实验、糖水消耗实验、悬尾实验和强迫游泳实验观察小鼠抑郁行为,高效液相色谱法测定小鼠前额叶皮层单胺类神经递质的水平。结果:行为学检测结果显示补阴消郁汤配方颗粒6g/kg、18g/kg剂量组能明显改善更年期抑郁模型小鼠行为,明显增加小鼠在旷场中央站立次数以及糖水消耗量,并且显著减少小鼠悬尾不动时间和游泳不动时间。脑内单胺类神经递质检测结果显示,补阴消郁汤配方颗粒18g/kg剂量组能明显增加5-HT和DA含量。结论:补阴消郁汤配方颗粒预防给药可以改善更年期模型小鼠的抑郁行为,其机制可能与调节小鼠脑内单胺类神经递质的含量有关。

Objective： To explore the protect effect of Buyin Xiaoyu Tang on ovariectomized model mice and its possible mechanism.Methods：Depression model by ovariectomized surgery was established after 4 weeks administration.we used open field test,sugar consumption test,tail suspension and forced swimming tests to explore the antidepressant effects of Buyin Xiaoyu Tang.Meanwhile,the monoamine neurotransmitter in the mouse brain was also determined by HPLC.Results： The results showed that Buyin Xiaoyu Tang significantly improved mice behavior at the dose of 6 g/kg and 18 g/kg dose groups（ P 〈 0.05）.Compared to the depression model group,the 5-HT and DA level were significantly increased in Buyin Xiaoyu Tang at 18 g/kg group（ P 〈 0.05）.Conclusions： These findings suggest that Buyin Xiaoyu Tang exerted antidepressant effect in despair tests and the underlying mechanism may involve the central monoamine neurotransmitter systems.
