摘要
药用野生稻积累了丰富的遗传多样性和有利基因,是水稻遗传改良的重要基础资源。LEA蛋白在植物抗逆反应起着重要的作用。本研究在药用野生稻干旱胁迫差异转录谱数据分析基础上,利用RT-PCR方法从药用野生稻克隆了OoLEA1基因,OoLEA1的开放阅读框长度为603 bp,编码一个由200个氨基酸残基组成的蛋白,分子量为20.73 k D,理论等电点为6.61。OoLEA1蛋白含有4个LEA_4结构域、8个α-螺旋构象和1个β-片层构象,该蛋白为亲水蛋白。OoLEA1受干旱和高温诱导表达,说明OoLEA1可能在水稻抗旱和抗高盐胁迫中发挥作用。
Oryza officinalis accumulates great genetic diversity and elite genes, and it is an important genetic source for rice improvement. LEA proteins played important roles in response to abiotic stress in plants. Based on the analysis of differently-expressed genes from drought-induced transcriptome of Oryza officinalis, the OoLEA 1 was cloned by RT-PCR technique. The opening read frame of OoLEA 1 was 603 bp length and was predicted to code a protein containing 200 amino acids with a molecular weight of 20.73 kD and a theoretical pI of 6.61. The OoLEA1 protein contained four LEA_4 domain, eight a-helical domains and only one β-sheet domain, which showed it was strongly hydrophilic. The expression of OoLEA 1 was induced significantly under the drought and high temperature stress, respectively. The results indicated OoLEA 1 might exert roles in resistance to drought and high temperature stresses.
