摘要
目的研究纳米颗粒二氧化钛(TiO_2NPs)和重金属Cd复合污染对水产食品的生态危害。方法采用光学立体解剖显微镜观察纳米颗粒TiO_2和重金属Cd复合污染处理后斑马鱼的形态变化,记录染毒48 hpf后的孵化数和染毒96 hpf后的死亡数,并且利用DCFH-DA探针检测了活性氧含量的变化。结果与仅TiO_2NPs污染相比较,TiO_2NPs和重金属Cd复合染毒48 hpf后,胚胎发生心包水肿,卵黄囊肿。染毒72 hpf后,胚胎外包偏少,并且斑马鱼胚胎的孵化率下降,死亡率上升,ROS含量升高。结论本研究可为水产食品业生物安全评价提供参考。
Objective To study the ecological harm of the combined pollution of nanometer particles TiO2 and heavy metals Cd in aquatic products. Methods An optical three-dimensional anatomical microscope was used to observe the morphology changes of zebrafish polluted by nano particles TiO2 combined with heavy metal Cd. And the hatching rate contaminated for 48 hpfand the mortality contaminated for 96 hpf were recorded, and the reactive oxygen species (ROS) content changes were detected by using DCFH-DA probe. Results Compared with only nanometer TiO2 particles pollution, embryogenesis pericardium edema and yolk cyst were formed in zebrafish after infected with composite nanometer particles TiO2 and heavy metals Cd for 48 hpf. After infected for 72 hpf, fewer embryos outsourcing occurred, the hatching rate of zebrafish embryo declined, the death rates increased, and the ROS levels increased. Conclusion This research can provide references for evaluation of biological safety of aquatic food.
